Constitutive activation of NF-κB inducing kinase (NIK) in the mesenchymal lineage using Osterix (Sp7)- or Fibroblast-specific protein 1 (S100a4)-Cre drives spontaneous soft tissue sarcoma

Abstract: Aberrant NF-κB signaling fuels tumor growth in multiple human cancer types including both hematologic and solid malignancies. Chronic elevated alternative NF-κB signaling can be modeled in transgenic mice upon activation of a conditional NF-κB-inducing kinase (NIK) allele lacking the regulatory TRAF3 binding domain (NT3). Here, we report that expression of NT3 in the mesenchymal lineage with Osterix (Osx/Sp7)-Cre or Fibroblast-Specific Protein 1 (FSP1)-Cre caused subcutaneous, soft tissue tumors. These tumors … Show more

“…In tumor bearing mice, Osterix , as well as the Osx-Cre allele, is also expressed in a subset of cancer-associated fibroblasts with a dual fibroblast/osteogenic signature 27 . Recently, we found subcutaneous sarcomas, but not bone tumors, in mice expressing a transgene driven by Osterix-Cre 33 . Thus, it is possible that loss of IKKα in adipocytes themselves is responsible for the observed fat phenotype.…”

“…Pretreatment with a specific inhibitor impeded the MHC-instigated toxicity in the MCF-7 cells, indicating caspase-3 activation during the MHC-induced apoptosis. It is now an established notion that an NF-κB over-activation is related positively with several human cancers [33]. Moreover, it also plays an important role in regulating the transcriptional activation of anti-apoptotic genes [34].…”

“…Large and cystic tumor masses were detected in the submandibular region, on the trunk, perineal area, and rarely on the limbs, with a predominance in females. Development of soft tissue tumors is related to constitutive activation of the alternative NF-κB pathway, including RelB in mesenchymal lineage cells [ 94 ].…”
