Construction and preliminary characterization of a river buffalo bacterial artificial chromosome library

Abstract: ABSTRACT. River buffalo genome analyses have advanced significantly in the last decade, and the genome sequence of Bubalus bubalis will be available shortly. Nonetheless, large-insert DNA library resources such as bacterial artificial chromosomes (BAC) are still required for validation and accurate assembly of the genome sequence. We constructed a river buffalo BAC library containing 52,224 clones with an average insert size of 97 kb, representing 1.7 × coverage of the genome. This genomic resource for river b… Show more

“…The river buffalo BAC library was three-dimensionally screened by PCR as described by Stafuzza et al (2012) using the following primers: forward 5ʹ-TGGCCCTCTCAGACCTGTTG-3ʹ and reverse 5ʹ-TGGCGAGGAGACTGAACACG-3ʹ (Amaral et al, 2008).…”

“…Genomic analysis of this species has advanced significantly in the last decade with the development of methodologies such as radiation hybrid panels for whole genome mapping (Amaral et al, 2007;Stafuzza et al, 2009) and genomic BAC libraries (Stafuzza et al, 2012) for region-specific resequencing and characterization of target genes (Stafuzza et al, 2014a,b,c).…”

“…Genome analysis methods have advanced significantly in the last decade and now include genomic tools such as the whole genome radiation hybrid panel used for genome mapping (Amaral et al, 2007(Amaral et al, , 2008 and the construction of a Murrah breed BAC library for resequencing of specific regions from the genome, as well as the development of high-density physical maps (Stafuzza et al, 2012). Recently, the genome sequence of the Mediterranean breed was publicly released by the National Center for Biotechnology Information database (NCBI -http://www.ncbi.nlm.nih.gov/assembly/67671).…”

“…Recently, another genomic tool, a buffalo large-insert bacterial artificial chromosome (BAC) library, was developed for the whole buffalo genome. This library facilitated the validation and accurate assembly of genome sequences, isolation of genes and gene clusters, elucidation of gene organization, and identification of regulatory and repetitive elements (Stafuzza et al, 2012).…”
