Construction of a BAC library of Korean ginseng and initial analysis of BAC-end sequences

Abstract: We estimated the genome size of Korean ginseng (Panax ginseng C.A. Meyer), a medicinal herb, constructed a HindIII BAC library, and analyzed BAC-end sequences to provide an initial characterization of the library. The 1C nuclear DNA content of Korean ginseng was estimated to be 3.33 pg (3.12 x 10(3) Mb). The BAC library consists of 106,368 clones with an average size of 98.61 kb, amounting to 3.34 genome equivalents. Sequencing of 2167 BAC clones generated 2492 BAC-end sequences with an average length of 400 b… Show more

“…The size of the ginseng genome is estimated to be 3.12 Gbp per haploid chromosome equivalent (Hong et al, 2004), whereas American ginseng, which recently diverged from ginseng , has an estimated genome size of 4.91 Gbp (Obae and West, 2012). However, details of the ginseng genome structure remain unknown due to the lack of genomic sequence data.…”

“…Research on ginseng has historically focused mainly on its medicinal components and their efficacy (Shibata et al, 1966;Lee and Huemer, 1971;Coon and Ernst, 2002;Park et al, 2012a). Genetic and genomic studies, such as functional analyses of genes (Tansakul et al, 2006;Kim et al, 2008Kim et al, , 2011, marker development Jo et al, 2011;Sun et al, 2011;Kim et al, 2012), construction of DNA libraries (Hong et al, 2004;Bang et al, 2010) and transcriptome sequencing (Chen et al, 2011;Wu et al, 2012) have been initiated only recently.…”

“…ginseng has an estimated genome size of over 3.5 Gbp for haploid genome equivalent (Hong et al 2004;Waminal et al 2012) and is regarded as an allotetraploid (2n=4x=48) . A previous study illustrated that P. ginseng genome had experienced two rounds of whole genome duplication (WGD) , and recent WGD occurring 2-3 million years ago (MYA), which made its genome much bigger than those of other diploid Panax species (Yi et al 2004;Choi et al 2013).…”
