Construction of a molecular genetic map for hawthorn based on SRAP markers

Abstract: The F1 hybrid population was a result from the crossing of 'Damianqiu' and 'Qiujinxing'. 'Damianqiu' was the female parent and 'Qiujinxing' was the male parent. Ninety-two F1 individual plants and their parents, in total of 94 materials, were chosen for the sequence-related amplified polymorphism analysis. The Joinmap3.0 software was used to construct the molecular genetic map of 'Damianqiu' and 'Qiujinxing', respectively. According to the result, the molecular linkage map of 'Damianqiu' has 21 linkage groups … Show more

“…SRAPs are dominant markers in nature. Generation of SRAP data and its analysis reveal valuable information for assessing genetic diversity of large germplasm collections and identification of quantitative trait loci in different taxa ( Wanga et al., 2015 ). As an ORF-based marker that targets functional genes, SRAP is very valuable in marker assisted crop breeding and improvement ( Bhatt et al., 2017 ; Ge and Daizhen 2015 ).…”

“…To date, there are only two reports on genetic map construction in hawthorn. Wang et al [23] first constructed the genetic maps for hawthorn cultivars 'Damianqiu' and 'Qiujinxing' by using SRAP markers and 92 hybrid offspring. However, the number of markers in these genetic maps was very low, and the average genetic distance between adjacent markers was very large, which was not suitable for fine QTL mapping.…”

“…However, the number of markers in these genetic maps was very low, and the average genetic distance between adjacent markers was very large, which was not suitable for fine QTL mapping. Furthermore, SRAP markers also showed poor repeatability and stability in linkage map construction [23] and not considerable for constructing genetic maps of other hawthorn cultivars. Zhao et al constructed other genetic maps by using 107 hybrid offspring of the same population and 2b-RAD technology and a total of 3894 SNP markers were achieved in the integrated map [24].…”

“…However, the relatively insufficient genetic mapping limits the molecular-assisted breeding of hawthorn to some extent. Wang G. et al (2015) constructed a rough genetic map of hawthorn using SRAP markers, with fewer than 100 markers and an average distance of more than 5 cM. Wang (2016) constructed a genetic map of hawthorn plants by using both SRAP and SSR markers, and the marker distance of male and female parents were 7.05 cM and 6.45 cM, respectively.…”
