Construction of a river buffalo (Bubalus bubalis) whole‐genome radiation hybrid panel and preliminary RH mapping of chromosomes 3 and 10

Abstract: ABSTRACT:The buffalo (Bubalus bubalis) not only is a useful source of milk, it also provides meat and works as a natural source of labor and biogas. To establish a project for buffalo genome mapping a 5,000-rad whole genome radiation hybrid panel was constructed for river buffalo and used to build preliminary RH maps from two chromosomes (BBU 3 and BBU10). The preliminary maps contain 66 markers, including coding genes, cattle ESTs and microsatellite loci. The RH maps presented here are the starting point for … Show more

“…Each biarmed water buffalo chromosome is derived from the fusion of two bovine acrocentrics. (a) This shows the similar banding patterns for bovine chromosomes 29 and 16 to water buffalo chromosome 5 22, ( b) This shows similar banding patterns for bovine chromosome 12 and water buffalo chromosome 13 22.…”

“…The RH panel for river buffalo is a recent development that has been used to construct preliminary RH maps for several of the water buffalo chromosomes 22-26. The preliminary RH maps for BBU1, BBU3, BBU6, BBU7, BBU10, and BBUX were based on markers derived from cattle, and showed that the bovine genome is a useful source of markers for buffalo genome mapping.…”

“…Myosin, heavy chain 7, cardiac muscle, beta (Amaral et al 2007;Goldammer et al 2007;Strafuzza et al 2007;Ianella et al 2008). Cytogenetic studies on female river buffaloes with reproductive problems (lack of oestrus or pregnancy, even in the presence of the bull) have revealed a high frequency (20%) of sterile females carrying sex chromosome abnormalities (reviewed in Di Meo et al 2008).…”

“…Genomic analysis of this species has advanced significantly in the last decade with the development of methodologies such as radiation hybrid panels for whole genome mapping (Amaral et al, 2007;Stafuzza et al, 2009) and genomic BAC libraries (Stafuzza et al, 2012) for region-specific resequencing and characterization of target genes (Stafuzza et al, 2014a,b,c).…”
