Construction of cavernosum smooth muscle using umbilical artery smooth muscle cells seeded on acellular corporal collagen matrices

Abstract: The objective of this study was to investigate the feasibility of tissue engineering of corpus cavernosal smooth muscle. Acellular corporal collagen matrices (ACCMs) were obtained from the penis of adult rabbits by a cell removal procedure. ACCMs were implanted into the back muscles of allogenic rabbits to investigate the resulting immunological reaction. Human umbilical artery smooth muscle cells (HUASMCs) were isolated from human umbilical arteries through explant techniques and expanded in vitro. Subsequent… Show more

“…Kershen et al (2002) Acellular corporal collagen matrices (ACCM) were used instead of PGA because of its unique 3D architecture. Reconstruction of CC using cells seeded on ACCM was investigated in five studies Eberli et al, 2008;Falke et al, 2003;Kwon et al, 2002;Song et al, 2009). Kwon et al (2002) harvested rabbit corporal tissue, which was processed to form acellular corporal tissue matrices ex vivo.…”

“…Song et al [19], citing recognized diffi culty in culturing pure cavernosal smooth muscle cells, investigated the potential of human umbilical artery smooth muscle cells for cavernosal regeneration. The authors successfully expanded the cells in culture and seeded the cells onto rabbit corporal cavernosal acellular grafts.…”

“…Another group seeded acellular corporal collagen matrices with SMCs derived from human umbilical arteries and implanted the cellularized implants into athymic mice. These authors reported that the scaffolds became cellularized to develop tissue with anatomical and functional properties similar to corporus cavernosum smooth muscle [ 150 ] .…”
