Contrast-enhanced Ultrasound in evaluating of angiogenesis and tumor staging of nasopharyngeal carcinoma in nude mice

Abstract: Objective To explore the use of Contrast-enhanced Ultrasound (CEUS) in evaluating angiogenesis in a xenograft nasopharyngeal carcinoma (NPC) model in nude mice and the evolution of CEUS parameters according to the growth of NPC. Methods Nude mice were divided into three groups according to experiments conducted at various times from tumor implantation (8 mice/group; group A: 4 weeks from implantation; group B:6 weeks from implantation; group C:8 weeks from implantation). CNE-2 cells were transplanted in 24 nud… Show more

“…Angiosuppression has been reported to parallel a decrease in tumor weight, as demonstrated in a rat gliosarcoma model upon copper chelation [ 46 ]. Moreover, the tumor volumes revealed a positive correlation with the microvessel density in a nasopharyngeal carcinoma mouse model [ 47 ]. Altogether, the observed decrease in the H460 tumor weight under low- and high-dose PSP-2 treatment, with concurrent significant angiosuppression, points towards an anti-cancer potential of PSP-2 via an anti-angiogenesis mechanism.…”

“…Contrast-enhanced ultrasound (CEUS) is an imaging technique that assesses tumor vasculogenesis by examining the diffusion patterns of contrast agents within a tumor. It has been claimed that MVD in NPC grafts can be reflected by CEUS parameters in a nude mouse model ( 95 ). Additionally, dynamic contrast-enhanced ultrasonography (DCE-US) can be used to evaluate the efficacy of anti-angiogenic therapy in NPC ( 96 ).…”

“…Once the sequence has been saved, either in-house software or contrast-specific software developed by the manufacturers can be used to map the intensity of the backscattered signal within the regions-of-interest (eg medulla, cortex) to study the perfusion-dynamics. Metrics of interest include area-under-the curve, time-to-peak enhancement, washin rate and these can be used as indicators of blood volume and vessel density [20]. In many instances if an ischemic-reperfusioninjury (IRI) mouse model is being studied, one kidney can act as a control and both B-mode and contrast-enhancement data can then be acquired from both kidneys.…”
