Contribution of spermatozoal centrosomes to the microtubule-organizing centre in Antarctic minke whale (Balaenoptera bonaerensis)

Abstract: SummaryUsing an interspecies microinsemination assay with bovine oocytes, it was examined whether centrosomes of Antarctic minke whale spermatozoa function as the microtubule-organizing centre (MTOC). Bull and rat spermatozoa were used as positive and negative controls, respectively. Vitrified-warmed bovine mature oocytes were subjected to immunostaining against α-tubulin 4-6 h after intracytoplasmic injection (ICSI) of 5 mM dithiothreitol-treated spermatozoa. Aster formation occurred from whale spermatozoa (3… Show more

“…To assess MTOC function in fertilized oocytes, FD-ICSI, control ICSI and IVF oocytes were subjected to immunostaining [32]. For FD-ICSI and control ICSI, injected oocytes were activated with ionomycin and ethanol at a 4-h interval and further cultured in TCM199+5% FBS for 3 h at 38.5 C under 5% CO2 in air.…”

“…The length of the comet tail (pixel) was measured from the center of gravity of the head, and the DNA content in the comet tail was determined with the relative fluorescent intensity and defined as % DNA liberated. The DNA fragmentation index (Moment) was calculated as the length of the comet tail × the % DNA liberated. To assess MTOC function in fertilized oocytes, FD-ICSI, control ICSI and IVF oocytes were subjected to immunostaining [32]. “…Assembly of the microtubule network is promoted by oocyte activation. The ratio of aster diameter to oocyte diameter is 0.57 in injected and activated oocytes (Kobayashi et al ., 2006).…”

“…This seems very promising especially when bovine embryos have been already produced with ICSI using freeze-dried spermatozoa, heat-dried sperm heads or frozen-thawed oocytes (Keskintepe et al, 2002, Rho et al, 2004Lee & Niwa, 2006). Interspecies microfertilization using bovine oocytes can also be used to study key processes during sperm-oocyte fusion, oocyte activation and fertilization (Li et al, 2003;Kobayashi et al, 2006). This has a special relevance in human fertility, as bovine ICSI has been suggested to be an appropriate model to assess human sperm oocyte activation ability (Terada et al, 2004).…”
