Contributions of PARP‐1 rs1136410 C>T polymorphism to the development of cancer

Abstract: According to the Global Cancer Statistics 2018, about 18.1 million new cancer cases and 9.6 million cancer deaths occur worldwide in 2018. 1 Single nucleotide polymorphisms (SNPs), the most frequent type of genetic alterations, have been proved to contribute to cancer susceptibility. 2 Yet, all the identified SNPs only account for a small portion of cancer risk. Poly(ADP-ribose) polymerases (PARP), also known as ADPribosyl-transferase diphtheria toxin-like (ARTDs), are evolutionary-conserved family of proteins… Show more

“…PARP1 polymorphism Val762Ala (rs1136410 T>C) may be associated with prostate cancer (106), esophageal squamous cell carcinoma (107), and breast cancer (108). Recent meta-analyses found a borderline significant association between PARP1 Val762Ala polymorphism and overall cancer risk, although after stratification by cancer types, the polymorphism could predispose to gastric cancer, thyroid cancer and cervical cancer, in an Asian population, but not in Caucasian and African populations (109,110). Involuntary smoking was associated with an increased risk of breast cancer among both preand postmenopausal women, depending on the genotype of PARP1 762 (111), while Ala762Ala (rs1136410 C/C) genotype was associated with an elevated risk of esophageal squamous cell carcinoma in smokers compared to T/T or T/C genotype (107,112).…”

“…In a meta-analysis, Li et al collected research results on PARP1 polymorphisms [ 88 ]. The authors showed that the rs1136410 polymorphism has a significant association with the overall risk of cancer, and detailed subgroup analyses showed that it may significantly predispose to stomach cancer, thyroid cancer, and cervical cancer while protecting against brain cancer.…”

“…This SNP has been linked to both lung cancer and follicular lymphoma through the HyDn-SNP-S method (2,3). The rs1136410 SNP has also been shown to serve as a protective factor against breast cancer and coronary artery disease in the Han Chinese population, but it may lead to an increased overall risk of age-related cataracts and cancers (61)(62)(63)(64)(65).…”
