Contriving Multi-Epitope Subunit of Vaccine for COVID-19: Immunoinformatics Approaches

Abstract: COVID-19 has recently become the most serious threat to public health, and its prevalence has been increasing at an alarming rate. The incubation period for the virus is ∼1-14 days and all age groups may be susceptible to a fatality rate of about 5.9%. COVID-19 is caused by a novel single-stranded, positive (+) sense RNA beta coronavirus. The development of a vaccine for SARS-CoV-2 is an urgent need worldwide. Immunoinformatics approaches are both cost-effective and convenient, as in silico predictions can red… Show more

“…Apart from this, β-defensin interact with the immune receptors such as TLRs or CCR6 and induces innate and adaptive immune responses (Lei et al., 2019 ). It is used as an adjuvant in the construction of several vaccines by Immunoinformatics approach (Ali et al., 2017 ; Bhatnager et al., 2020 ; Dong et al., 2020 ; Narula et al., 2018 ).…”

“…Apart from this, b-defensin interact with the immune receptors such as TLRs or CCR6 and induces innate and adaptive immune responses (Lei et al, 2019). It is used as an adjuvant in the construction of several vaccines by Immunoinformatics approach (Ali et al, 2017;Bhatnager et al, 2020;Dong et al, 2020;Narula et al, 2018). Apart from b-defensin, PADRE (pan DR epitope) sequence was also included in the vaccine construct to increase the immunogenicity of the vaccine construct as well as to enhance the T-cell help (Alexander et al, 2000;Sarkar et al, 2020;Wu et al, 2010).…”

“…Effective preventive approaches are urgent needs at the current situation. Potent epitopic vaccines predicted by bioinformatic analysis makes the vaccine design straightforward and fast compared to traditional vaccine approaches, which has been used in COVID-19 vaccine design recently [ 35 , 63 65 ].…”

“…In addition, structural proteins (surface glycoprotein, envelope protein, and membrane glycoprotein) of SARS-CoV-2 were selected from GenBank, and several immunoinformatics coupled with computational approaches were employed to forecast B- and T-cell epitopes from the SARS-CoV-2 highly antigenic structural proteins to design an effective MESV (Tahir ul Qamar et al, 2020b ). Another study to design a multiepitope vaccine, retrieving 27 reference sequences of SARS-CoV-2 proteins from the National Center for Biotechnology Information (NCBI) Protein Database ( https://www.ncbi.nlm.nih.gov/protein ), selecting proteins with ≥100aa and an antigenic score of ≥0.5 for further structural modeling (Dong R. et al, 2020 ).…”
