Control of Particle Size by Coagulation of Novel Condensation Aerosols in Reservoir Chambers

Abstract: The coagulation growth behavior of capillary aerosol generator (CAG) condensation aerosols was investigated in a series of reservoir chambers. Aerosols consisted of a condensed system of 0.7% w/w benzil (model drug) in propylene glycol (vehicle). These were generated into 250-, 500-, 1,000-, and 2,000-mL reservoirs in both flowing air-stream and static air experiments. Changes in drug and total aerosol particle size were measured by a MOUDI cascade impactor. In both series of experiments the CAG aerosols grew … Show more

“…It is well known that cigarette smoke contains primarily particles of mass median aerodynamic diameter near 0.6 m (Porstendorfer and Schraub, 1972), outside of the desirable 1-to 3-m size range. Previously, Hong et al (2002) studied the relationship between condensation aerosol particle size and mixing of vapor into air, using a drug surrogate dissolved into solvent as the model system. They found a trend, consistent with the one shown in Fig.…”

“…A subsequent increase in droplet size occurs due to condensation of the surrounding vapour onto the formed nuclei, generating the desired aerosol for pulmonary delivery . The appropriate droplet size can be achieved by controlling droplet coagulation using reservoir chambers …”

“…As seen with Respimat ® , this type of nebulizer has been used to produce submicron droplets to investigate the concept of enhance excipient growth . Importantly, by dissolving benzyl in propylene glycol, it has been shown that both evaporate and condensate simultaneously . The aerosol droplet is also dependent on energy input, with trough in MMAD at about 40 Joules .…”

“…The condensation of surrounding vapor onto the generated nuclei results in an aerosol. Various drugs, such as perphenazine, 173 prochlorperazine, 174 rizatriptan 175 and benzyl, 176 have been aerosolized using this technique. Propylene glycol (PG) is a popular solvent chosen to dissolve the drugs.…”
