Control of Protein Homeostasis in the Early Secretory Pathway: Current Status and Challenges

Abstract: : Discrimination between properly folded proteins and those that do not reach this state is necessary for cells to achieve functionality. Eukaryotic cells have evolved several mechanisms to ensure secretory protein quality control, which allows efficiency and fidelity in protein production. Among the actors involved in such process, both endoplasmic reticulum (ER) and the Golgi complex play prominent roles in protein synthesis, biogenesis and secretion. ER and Golgi functions ensure that only properly folded p… Show more

“…Macroautophagy (herein referred to as autophagy) is a cytoprotective lysosomal pathway for the degradation of superfluous or potentially dangerous cytosolic material and organelles. [173][174][175][176] Robust evidence indicates that autophagy is required for the preservation of lysosomal ATP stores in the course of most (but not all) variants of ICD. 149 166 As a consequence of cellular blebbing, which is mediated by lysosomal-associated membrane protein 1 (LAMP1) and pannexin 1 (PANX1), ATP is released into the extracellular space.…”

“…Thus, they belong to that part of the cell proteome translocating into the endoplasmic reticulum (ER) for maturation and eventually entering the secretory pathway to reach the cell surface [9]. In this compartment, a stringent quality control system scrutinizes the newly synthetized proteins in order to: (i) allow the properly folded polypeptides to pass forward and (ii) deliver to degradation those folding-defective [36,37]. This ensures protein homeostasis avoiding, at the same time, the accumulation of potentially harmful species.…”

“…About one-third of the eukaryotic cellular proteome is folded and matured in the endoplasmic reticulum (ER) ( Sicari et al., 2019 ). Often, proteins fail to fold or assemble properly and are eliminated by the ER-associated degradation (ERAD) pathway ( Sun and Brodsky, 2019 ).…”
