Control of RNA Stability by NrrF, an Iron-Regulated Small RNA in Neisseria gonorrhoeae

Abstract: bRegulation of gene expression by small noncoding RNAs (sRNAs) plays a critical role in bacterial response to physiological stresses. NrrF, a trans-acting sRNA in Neisseria meningitidis and Neisseria gonorrhoeae, has been shown in the meningococcus to control indirectly, in response to iron (Fe) availability, the transcription of genes encoding subunits of succinate dehydrogenase, a Fe-requiring enzyme. Given that in other organisms, sRNAs target multiple mRNAs to control gene expression, we used a global appr… Show more

“…MtrF is also by itself a proton motive force (PMF)-dependent antibiotic efflux pump that expels sulfonamides from the bacteria (32). Interestingly, trans-acting, iron-regulated sRNA NrrF directly controls MtrF expression by reducing mtrF mRNA stability by increasing its turnover (33,58). Thus, NrrF attenuates MtrF action in antibiotic resistance.…”

“…Thus, NrrF attenuates MtrF action in antibiotic resistance. MtrF transcripts are also repressed by Fur, MtrR (repressor of mtrCDE), and MpeR (repressor of mtrR) (33), implying that expression of that inner membrane protein is tightly controlled by several additional regulators.…”

“…Fe-deplete (CDM-0) and Fe-replete medium (CDM-10), supplemented with 10 μm Fe (NO 3 ) 3 were prepared as previously reported [16, 52]. Briefly, organisms were inoculated to an OD 600  = 0.100 from overnight growth on GCB agar plates.…”

“…The latter binds and thereby represses tolC mRNA in E. coli and Salmonella [155]. Other examples include MdtEF regulation by DsrA in E. coli [156] and MtrF (part of MtrCDE) regulation by NrrF in Neisseria gonorrhoeae [157]. Furthermore, sRNAs can 're-wire' two component cascades and therewith indirectly influence expression level [158].…”
