Controlled comparison of the efficacy and safety of four doses of suriclone, diazepam, and placebo in generalized anxiety disorder

Abstract: The anxiolytic activity and tolerance of four doses of suriclone (0.1, 0.2, 0.3 and 0.4 mg tid), diazepam (5 mg tid), and placebo were compared in six parallel groups of 54-59 outpatients with generalized anxiety disorder (DSM III-R). After a 1-week placebo run-in period, the patients were treated for 4 weeks, with assessments at baseline and after 1, 2, and 4 weeks by the Hamilton anxiety scale and the Clinical Global Impressions. Results showed better improvement with active drugs as compared to placebo, wit… Show more

“…At these doses, suriclone exhibited adverse effects different from those of benzodiazepines, with more dizziness and less sedation. In a recent study on the efficacy and safety of 0.1, 0.2, 0.3 and 0.4 mg suriclone and 5 mg diazepam, these drugs demonstrated anxiolytic effects when compared to placebo without interdrug differences, but suriclone was better tolerated than diazepam (Ansseau et al 1991).…”

“…Á Diazepam was effective in studies containing a placebo condition (Ansseau et al 1991;Boyer and Feighner, 1993;Fontaine et al 1983;Rickels et al 1993Rickels et al , 1997Rickels et al , 2000a as well as studies using a comparator with established efficacy ( Pregabalin. Pregabalin was superior to placebo in a placebo-controlled study (Pohl et al 2005).…”

“…Dose-dependent toxic effects were dizziness and ataxia. In comparative clinical trials with patients exhibiting generalized anxiety disorders suriclone (0.3-2 mg day-) proved to be as effective as diazepam (15-25 mg day-) [6,7] and lorazepam (5 mg day-') [8]. The somatic complaints profile was different with suriclone compared with diazepam: alertness was less influenced but dizziness appeared to be more frequent after suriclone.…”
