Controlled Human Malaria Infection of Healthy Adults With Lifelong Malaria Exposure to Assess Safety, Immunogenicity, and Efficacy of the Asexual Blood Stage Malaria Vaccine Candidate GMZ2

Abstract: Background GMZ2 is a recombinant malaria vaccine inducing immune responses against Plasmodium falciparum (Pf) merozoite surface protein-3 and glutamate-rich protein. We used standardized controlled human malaria infection (CHMI) to assess the efficacy of this asexual blood-stage vaccine. Methods We vaccinated 50 healthy, adult volunteers with lifelong exposure to Pf 3 times, at 4-week intervals, with 30 or 100 µg GMZ2 formula… Show more

“…This substantially contributed to lower VE calculations in both arms of the study. Although 100% of nonimmune control subjects in the United States and Europe (73/73) 13,26,27,29,40,41 and Tanzania (34/34) 15,18 have been infected with this dose, in other settings in Africa, including Gabon (20/25), 27,42 Gambia (17/ 19), 28 and unpublished data from Mali (8/15) and Kenya (137/ 170) (submitted), this has not been the case.…”

“…We identified 5 Sub-Saharan African HCS involving falciparum malaria in Tanzania, Kenya, and Gabon (Hodgson et al 2014;Shekalaghe et al 2014;Lell et al 2017;Dejon-Agobe et al 2018;Jongo et al 2018). These studies are part of an international research program using cryopreserved P. falciparum (Pf) sporozoites from a laboratory strain (NF54) known to be sensitive to chloroquine and produced in increasingly controlled laboratory settings since the 1980s (Chulay et al 1986).…”
