The mechanism for the accelerating effects of epidermal growth factor (EGF) and insulin-like growth factor I (IGF-I) on the meiotic cell cycle of bovine oocytes cultured in vitro was investigated. Cumulus-oocyte complexes (COCs) were obtained from small (< or = 3 mm in diameter), medium (4-6 mm in diameter) or large (7-10 mm in diameter) ovarian follicles and cultured with or without a combination of EGF and IGF-I (growth factors). Growth factors significantly increased the frequency of first polar body extrusion of oocytes derived from small follicles at 16 h of culture (PB16 oocytes; with growth factors: 75%; without growth factors: 55%), but did not increase the frequency in oocytes from medium or large follicles. COCs from small follicles were cultured with individual growth factors and sampled for kinase activity. The frequencies of polar body extrusion in EGF only (67%) and EGF + IGF-I (75%) treatment groups were significantly higher than those in the control (no growth factor) group (49%), but not significantly higher than in the IGF-I only group (63%). The H1 kinase activity at 6-8 h of culture in each group increased significantly from the baseline value at 0 h of culture, and the H1 kinase activities in the EGF only, IGF-I only and EGF + IGF-I treatment groups were significantly higher than those in the control group at 8 h of culture. MAP kinase activity was significantly higher than the baseline value and significantly higher than that in the control group at 6 h of culture in the EGF treatment group only. In conclusion, EGF and IGF-I act on COCs from small follicles to accelerate the meiotic cell cycle of the oocytes. This accelerating effect may be related to increased H1 and MAP kinase activities during the early stages of maturation.

The objectives of this study were to evaluate: (1) the nuclear maturation, (2) the intracellular glutathione (GSH) content, (3) the normality of fertilization and (4) full development after transplantation of embryos derived from porcine oocytes precultured with 50 mmol/l roscovitine (an inhibitor of p34 cdc2 /cyclin B kinase) for 22 h. After treatment with roscovitine, the nuclear configuration of oocytes (Hoechst staining) was comparable with those examined just after collection: the majority of oocytes were arrested at the germinal vesicle (GV) 1 stage (63.2%). Roscovitine-treated oocytes progressed through meiosis to the metaphase II stage in a conventional step-wise in vitro maturation (IVM) program for 44 h in a proportion similar to control ones (> 85.0%). When roscovitine-treated oocytes and non-treated oocytes were matured for 44 h and then co-cultured with fresh spermatozoa for 18 h, no differences were observed in oocyte penetrability, proportion of monospermic penetration and male pronuclear formation (>87%). Roscovitine increased the GSH synthesis in oocytes at 22 h, whereas, after 44 h, roscovitine-treated oocytes had similar amounts of GSH to non-treated oocytes. Finally, surgical transfer of zygotes at 22-24 h post-insemination, derived from roscovitine-treated oocytes, resulted in one pregnancy with 12 piglets born; control non-treated zygotes resulted in one pregnancy and 10 piglets born. The full-term developmental ability of mammalian oocytes pre-cultured with roscovitine prior to IVM is thereby demonstrated. ResumoFoi avaliada a criotolerância de oócitos e embriões bovinos maturados com adição de líquido folicular (LF) e/ou β-mercaptoetanol (BM). Após vitrificação, os oócitos foram maturados em: TCM-199 (controle); BM (24h TCM-199+100µM BM); LF (6h em LF+18h TCM-199) e LF+BM (6h LF+18h TCM-199+100µM BM). Não houve diferença (p>0,05) nas taxas de blastocistos dos tratamentos TCM (6,4%), BM (4,0%) e LF (3,4%). A eclosão e densidade celular dos embriões eclodidos não diferiram (p>0,05) nos tratamentos. No Experimento 2 blastocistos expandidos (Bx) obtidos em D7 ou D8 foram vitrificados, avaliando-se sua reexpansão e eclosão. A reexpansão foi semelhante (p>0,05), sendo observado comportamento distinto na eclosão entre Bx D7 e D8. Nos Bx D7 houve maior eclosão no controle (TCM-54,2%) em relação ao BM (40,32%) e LF+BM (33,89%). Os Bx D8 apresentaram menor eclosão no controle (TCM) em relação aos Bx D7. Nos tratamentos BM, LF e LF+BM a eclosão foi semelhante para Bx D7 ou D8. A maturação com adição de LF e/ou BM não melhora a criotolerância de oócitos imaturos e embriões PIV. Blastocistos expandidos precoces (D7) são mais criotolerantes e apresentam um comportamento distinto à adição de LF e BM, em relação aos tardios (D8).
