Convergence of eicosanoid and integrin biology: Role of Src in 12-LOX activation

Abstract: Abstract12-Lipoxygenase (12-LOX) metabolizes arachidonic acid to 12(S)-hydroxyeicosatetraenoic acid, or 12(S)-HETE, a proinflammatory bioactive lipid implicated in tumor angiogenesis, growth, and metastasis. The mechanisms underlying 12-LOX-mediated signaling in cancer progression are still ill-defined. In the present study we demonstrate that 12-LOX phosphorylation and subsequent enzymatic activity occurs after integrin β4 stimulation and Src kinase recruitment to the integrin subunit. Inhibition of Src activ… Show more

“…Overexpression of the platelet 12-LOX in PC-3 prostate cancer cells results in 12(S)-HETE production which leads to the activation of transcription factors such as NF-κB resulting in increased MMP9 expression and in hypoxia inducible factor-1alpha (HIF-1alpha)-mediated in VEGF expression [ 71 , 72 , 117 ]. There is also evidence that platelet 12-LOX-mediated AA metabolism is responsible for radioresistance of prostate cancer cells and metastatic potential of melanoma cells [ 118 ].…”

“…In turn, these autonomously-generated gradients cause self-organization of the cells, such that they migrate as a group [ 46 , 47 ]. Although we have not detected differences in mRNA expression of the 12-S-HETE biosynthetic proteins, or the 12-S-HETE receptor (GPR31) between winner and loser GSCs, in human prostate cancer expression levels of GPR31 and 12-S-HETE both positively correlate with metastasis [ 48 , 49 ]. The link between relay signaling, clustering and competition is the subject of current studies.…”

“…12-LOX has been identified as a crucial player in diabetes, skin diseases, thrombosis and cancers 15,22–27. There is increasing evidence that 12-LOX and 12-HETE promote tumor angiogenesis, growth and metastasis 28. Genetic studies suggest that mutations of 12-LOX are closely associated with tumorigenesis 29.…”
