Correlation between the 1.6 Å crystal structure and mutational analysis of keratinocyte growth factor

Abstract: A comprehensive deletion, mutational, and structural analysis of the native recombinant keratinocyte growth factor (KGF) polypeptide has resulted in the identification of the amino acids responsible for its biological activity. One of these KGF mutants (A23KGF-Rl44Q) has biological activity comparable to the native protein, and its crystal structure was determined by the multiple isomorphous replacement plus anomalous scattering method (MIRAS). The structure of KGF reveals that it folds into a p-trefoil motif … Show more

“…In conclusion, the data presented in this report show that FGF4 adopts a typical ␤-trefoil fold similar to other FGFs (24,28,43). A ternary FGF4-FGFR1-heparin model constructed by superimposing FGF4 onto FGF2 in the FGF2-FGFR1-heparin structure assisted the identification of several key residues in FGF4 involved in receptor and heparin binding.…”

“…2a). The ␤4 strand and N terminus of FGF7 have been shown to be required for FGFR2b binding by prior biochemical studies (33,(36)(37)(38). Similarly, the ␤CЈ-␤E loop of the receptor has been shown to participate in FGF7 binding (39)(40)(41).…”

“…1a and 2a). Superimposition of FGF10 onto FGF7, for which the crystal structure only in the absence of the receptor is available (33,34), gives a rms deviation (rmsd) of 0.885 Å (Fig. 2b).…”

“…Members of the FGF family share between 10 and 55% sequence identity (6). Crystal structures of FGF1 (7), FGF2 (7)(8)(9), and FGF7 (10), have revealed a common core FGF structure consisting of three copies of a four-stranded ␤-sheet, known as a ␤-trefoil fold. All FGFs contain N-and C-terminal segments outside the ␤-trefoil core.…”
