Correlation of nitric oxide produced by an inducible nitric oxide synthase-like protein with enhanced expression of the phenylpropanoid pathway in Inonotus obliquus cocultured with Phellinus morii

Abstract: Fungal interspecific interactions enhance biosynthesis of phenylpropanoid metabolites (PM), and production of nitric oxide (NO) is known to be involved in this process. However, it remains unknown which signaling pathway(s) or regulator(s) mediate fungal PM biosynthesis. In this study, we cocultured two white-rot fungi, Inonotus obliquus and Phellinus morii, to examine NO production, expression of the genes involved in phenylpropanoid metabolism and accumulation of phenylpropanoid-derived polyphenols by I. obl… Show more

“…A rapid nitrosative burst following fungal recognition drives NO accumulation, which activates the transcription of genes in phenylpropanoid metabolism required for styrylpyrone polyphenol biosynthesis4 followed by S-nitrosylation of PAL, 4CL and SPS12. Our data further confirm that these key enzymes in the biosynthesis of these natural products are S-nitrosylated and this modification reduces their activity, decreasing production of styrylpyrone polyphenols.…”

“…The phenylpropanoid pathway is integral to styrylpyrone biosynthesis and PAL and 4CL are key enzymes for the production of phenylpropanoid related metabolites4. The regulation of the phenylpropanoid synthesis is principally thought to occur at a transcriptional level25.…”

“…Alternatively, increases in ROS levels may function as a defence signalling response similar to that in plants, for example, triggering biosynthesis of pigment (94,96). Increases in another potential signalling compound, nitric oxide (NO), have also been detected during interactions between Phellinus morii and Inonotus obliquus, triggering the production of antifungal phenylpropanoid metabolites (97).…”
