Cortical Microtubule Arrays Lose Uniform Alignment Between Cells and are Oryzalin Resistant in the Arabidopsis Mutant, radially swollen 6

Abstract: The coordinated expansion of cells is essential to the formation of correctly shaped plant tissues and organs. Members of the radially swollen (rsw) class of temperature-sensitive arabidopsis mutants were isolated in a screen for reduced anisotropic expansion, by selecting plants with radially swollen root tips. Here we describe rsw6, in which cortical microtubules in the root epidermis are well organized in parallel arrays within cells, but neighboring cells frequently contain arrays differing in their mean o… Show more

“…4B). Under higher magnification, this phenotype resembled the previously reported radially swollen mutants (19,20). No obvious shoot phenotype was detected, possibly based on the less severe RanGAP1 depletion in shoots.…”

“…Seedlings were grown for 5 d on 1% sucrose and then transferred onto plates containing 1% sucrose, 4.5% sucrose, or 1% sucrose plus 50 mM NaCl for 3 d. Seedlings were fixed, stained for microtubules, and imaged, all as described (Bannigan et al, 2006). Briefly, the fixative contained 4% paraformaldehyde, 1% glutaraldehyde, 50 mM PIPES, and 1 mM CaCl 2 .…”

“…The rsw4 line was described previously (Wiedemeier et al, 2002). The GFP-tubulin reporter line expresses GFP fused to the A. thaliana -tubulin 6 (TUB6) gene (Bannigan et al, 2006). The GFP-histone lines express GFP fused to the A. thaliana histone H3.3 gene (At5g10980) and were made by Keith Earley and Craig Pikaard (Washington University of St Louis).…”
