CRABP1 in Non-Canonical Activities of Retinoic Acid in Health and Diseases

Abstract: In this review, we discuss the emerging role of Cellular Retinoic Acid Binding Protein 1 (CRABP1) as a mediator of non-canonical activities of retinoic acid (RA) and relevance to human diseases. We first discuss the role of CRABP1 in regulating MAPK activities and its implication in stem cell proliferation, cancers, adipocyte health, and neuro-immune regulation. We then discuss an additional role of CRABP1 in regulating CaMKII activities, and its implication in heart and motor neuron diseases. Through molecula… Show more

“…Figure 1A shows the highly conserved (>99%) sequence of the Mus musculus CRABP1 isoform (Uniprot ID P62965), with the relevant secondary structures superimposed over the corresponding amino acid sequence. The only non-conserved residue is proline 86, which exists as an alanine in the human and bovine orthologs ( Figure 1A , asterisk) ( Nhieu et al, 2022 ). In this study, we examine residues within helix 2 (blue highlight) and beta strands 7 and 8 (yellow highlights) that all contribute to its activity in interacting with, and regulating, CaMKII ( Figure 1A ).…”

“…This resulted in the identification of a novel CRABP1 p.(Glu18Asp) variant present only in cases 301, 302 and their mother 201. Cellular retinoic acid–binding protein 1 ( CRABP1 ; *180230) is a tumor suppressor gene with an important role in retinoic acid‐mediated differentiation and proliferation, which has been associated with varying cancer types (Banz et al., 2010; Nhieu et al., 2022). It is implicated as a highly conserved gene across mammals, thus variations are hypothesized as potential inducers of tumor formation or progression (Nhieu et al., 2022).…”

However at this stage, it is difficult to draw a definite conclusion about the role of CRABP1 p.(Glu18Asp) variant in ESC onset, since there is only case 201 with a clinical diagnosis.…”

“…In both PFAs and DMGs, its expression correlated with specific tumor cell types, with an enrichment among progenitor-like cells. While its precise role remains unclear, CRABP1 has been proposed to be a critical regulator of retinoic acid signaling, preventing canonical RA- induced transcriptional programs [54]. This role is of particular interest due to retinoic acid’s function in hindbrain patterning [7, 23, 30, 47, 55] and the previous identification of the retinoic acid binding protein RXRA as the top ranked super-enhancer associated gene in H3K27M DMGs [52].…”
