CRISPRz: a database of zebrafish validated sgRNAs

Abstract: CRISPRz (http://research.nhgri.nih.gov/CRISPRz/) is a database of CRISPR/Cas9 target sequences that have been experimentally validated in zebrafish. Programmable RNA-guided CRISPR/Cas9 has recently emerged as a simple and efficient genome editing method in various cell types and organisms, including zebrafish. Because the technique is so easy and efficient in zebrafish, the most valuable asset is no longer a mutated fish (which has distribution challenges), but rather a CRISPR/Cas9 target sequence to the gene … Show more

“…We generated a genetically defined zebrafish strain to simplify target site selection 19 , a rapid assay (CRISPR-STAT) for determining the somatic activity of sgRNA 20 , and a public database to curate published sgRNA targets and activities 21 in the belief that the transfer of sgRNA information is faster and cheaper than transferring actual mutant zebrafish. We used our high-throughput mutagenesis pipeline to generate the largest published germline transmission dataset in zebrafish 17 .…”

“…Several resources have been established to address this need. These include CRISPRz (12), a database of sgRNAs validated in zebrafish, WGE (13), a data resource that contains information about sgRNAs that can be used to target genes of interest and CrisprGE (14), a platform that provides knowledge about sequence mutations caused by sgRNAs previously used in various experimental settings. Nevertheless, a database that allows to compare screening results, such as perturbation phenotypes or sgRNA efficiency, of many different experiments on a genome-wide scale has so far not been available.…”
