Critical role of NLRP3-caspase-1 pathway in age-dependent isoflurane-induced microglial inflammatory response and cognitive impairment

Abstract: BackgroundElderly patients are more likely to suffer from postoperative cognitive dysfunction (POCD) after surgery and anesthesia. Except for declined organ function, the particular pathogenesis of POCD in elderly patients remains unknown. This study is carried out to determine the critical role of the NOD-like receptor protein 3 (NLRP3)-caspase-1 pathway in isoflurane-induced cognitive impairment.MethodsYoung (6–8 months old) and aged (14 months old) healthy male C57BL/6 mice were exposed to 1.5% isoflurane f… Show more

“…17 The contents of IL-1, IL-6, and TNF-α in the patientʼs plasma will be increased, which are closely related to the degree of surgical trauma. 19 Thus, they will activate the neuroglial cells, and produce a series of immune reactions to produce excessive inflammatory substances (IL-1β, IL-6, TNF-α, interferon-γ, cyclooxygenase-2, and complement) in the central nervous system. 18 In the case of a peripheral inflammatory response, a large number of inflammatory factors will enter the brain tissue through various pathways.…”

“…The inflammasome has been reported to be related to the pathogenesis of PND in various murine models [16,69,77,78]. In a mouse model subjected to anesthesia (isoflurane), aged mice, but not young ones, showed activation of the NLRP3-caspase-1 pathway, poor performance in the Morris water maze, and increased levels of IL-1β and IL-18 [16].…”

In a mouse model subjected to anesthesia (isoflurane), aged mice, but not young ones, showed activation of the NLRP3-caspase-1 pathway, poor performance in the Morris water maze, and increased levels of IL-1β and IL-18 [16]. Moreover, in mice subjected to major abdominal surgery, components of the NLRP1 inflammasome complex are expressed and IL-1β and IL-18 are highly produced in the spleen, whereas the inhibition of the NF-κB p65 transcript reduced these effects [77].…”
