Cross-linking of T-cell receptors on double-positive thymocytes induces a cytokine-mediated stromal activation process linked to cell death.

Abstract: To investigate molecular events associated with the intrathymic process of negative selection, we established an in vivo system using an anti‐CD3 epsilon monoclonal antibody to induce synchronous apoptosis in the thymus of AND T‐cell receptor (TCR) transgenic RAG‐2−/− mice in a non‐selecting haplotype. This model eliminates endogenous negative selection as well as gene activation in the mature thymocyte compartment, offering an ideal source of tester (anti‐CD3 epsilon‐treated) and driver (untreated) thymus RNA… Show more

“…However, the high level of UBP43 expression in the thymus suggest a possible link between UBP43 and immature T-cell apoptosis. Therefore, we also analyzed UBP43 expression in a mouse model with regulated thymocyte apoptosis (28). There were no observed changes in UBP43 expression in thymic RNA derived from either untreated mice or mice treated with anti-CD3 ε monoclonal antibody to induce thymocyte apoptosis (data not shown).…”

“…anti-CD3 Ab, a phenomenon that is probably related to autoAginduced negative selection during T cell maturation (52)(53)(54)(55)(56)(57). Previously, it was noted that the CD4 ϩ CD8 ϩ thymocytes isolated from the transgenic mice that expressed a DNI-B␣ protein in T cells were protected from anti-CD3-induced apoptosis (37).…”

“…Crosslinking of TCRs on DP thymocytes induces the production of various cytokines (14) and numerous studies have shown that cytokines such as IL-7 and IL-2 can profoundly affect thymocyte growth and survival (15). However, the precise role of IL-2/IL-2R signaling in thymocyte development remains controversial due to apparently contradictory studies providing evidence for both the promotion and inhibition of thymocyte growth by this signaling pathway (16).…”
