The effects of the cryoprotectants dimethyl sulfoxide, glycerol and methyl glycol at different concentrations on Austrolebias minuano sperm quality parameters were evaluated in this study. The cellular kinetic parameters, determined using flow cytometry, indicated the best results with the samples cryopreserved with 7.5% methyl glycol. Dimethyl sulfoxide concentrations of 7.5% and glycerol concentrations of 10%, 12.5% and 15% demonstrated the least benefit across all evaluated sperm kinetic parameters. When assessed using flow cytometry, a concentration of 7.5% methyl glycol similarly showed better sperm kinetic parameters than 12.5% dimethyl sulfoxide. We conclude that cryoprotectants, especially methyl glycol, are effective for the preservation of sperm quality in A. minuano. However, high sensitivity of spermatozoa against glycerol was observed in these studies; thus, it is not recommended for cryopreservation purposes. The present study evaluated the effect of cryoprotectants, semen diluents and chicken lines during pellet method of semen cryopreservation. Three different experiments were conducted; Experiment 1 -semen was cryopreserved using dimethylformamide (DMF) at 6% and 9% concentrations in two semen diluents (Lake and Ravie diluent and TES/NaCl diluent), Experiment 2 -semen was cryopreserved using dimethylacetamide (DMA) at 6% and 9% with or without sucrose (100mM), Experiment 3-semen from two chicken lines (PD1 and PD6) was cryopreserved using DMA (6% and 9%). Semen was evaluated pre and post cryopreservation for progressive motility, live and abnormal sperm. Semen pellets were stored in cryovials for at least seven days before examination and insemination. Thawed semen was inseminated intravaginaly to study fertility. All the parameters studied were significantly lower (p<0.05) in cryopreserved semen. DMF in Lake and Ravie diluent gave very low fertility and TES/NaCl diluent no fertile eggs. DMA as cryoprotectant gave fertility up to 9.22 %. Addition of sucrose along with DMA produced fertility similar to other cryopreservation treatment groups. No difference in in vitro semen parameters between chicken lines was observed. There is difference in cryopreservation outcome due to semen diluent and type of cryoprotectant. This study was designed to test the fertilizing ability of cryopreserved turkey semen, and here, two experiments were performed: an in vitro analysis to assess the effects of Tselutin and Lake diluents and an in vivo test to determine the fertility and hatching rates by also studying the feat of three insemination doses (250, 400 and 600 × 106 sperm/hen). Pooled semen samples were diluted with Tselutin or Lake extender which contained 20% of dimethylsulfoxide and 1 mM of Ficoll at final sperm concentration of 3 × 109 sperm/mL. Thereafter, semen was packaged into straws and frozen on liquid nitrogen. The post-thaw sperm quality was evaluated considering motility (computer-aided sperm analysis—CASA system) and membrane integrity (flow cytometry). Significantly higher values of progressive motility and some kinetic parameters in semen frozen with Lake were found. When we compared the extenders in vivo, no significant effects were detected, whilst sperm concentration significantly affected both fertility and hatching rates, with the best results obtained with the sperm concentration of 400 × 106 sperm/hen. From the results obtained, it emerged that the extender type only affected sperm motility characteristics, not the fertilizing ability of frozen-thawed semen, while inseminating dose markedly affected fertility and hatching rates.
