Crystal cell rupture after injury in Drosophila requires the JNK pathway, small GTPases and the TNF homolog Eiger

Abstract: The prophenoloxidase-activating cascade is a key component of arthropod immunity. Drosophila prophenoloxidase is stored in crystal cells, a specialized class of blood cells from which it is released through cell rupture. Within minutes after bleeding, prophenoloxidase is activated leading to visible melanization of the clot matrix. Using crystal cell rupture and melanization as readouts to screen mutants in signal transduction pathways, we show that prophenoloxidase release requires Jun N-terminal kinase, smal… Show more

“…In addition, the release of PPOs is affected by mutations in small Rho GTPases, supporting a role for the cytoskeleton in crystal cell rupture. 21 Although PO is viewed as a key enzyme in melanin biosynthesis, other enzymes may play important roles in synthesizing melanin during melanization (Fig. 2).…”

“…An additional potential role for melanin production at the wound site involves the formation of reactive oxygen species and by-products of melanin production that have cytotoxic antimicrobial properties (58). Interestingly, the JNK cascade is necessary for crystal cell rupture following injury, which leads to melanization at the wound site (4).…”
