Crystal Structure of Hexokinase KlHxk1 of Kluyveromyces lactis

Abstract: Crystal structures of the unique hexokinase KlHxk1 of the yeast Kluyveromyces lactis were determined using eight independent crystal forms. In five crystal forms, a symmetrical ringshaped homodimer was observed, corresponding to the physiological dimer existing in solution as shown by small-angle x-ray scattering. The dimer has a head-to-tail arrangement such that the small domain of one subunit interacts with the large domain of the other subunit. Dimer formation requires favorable interactions of the 15 N-te… Show more

“…An extensive hydrogen-bonding network was observed between the hydroxyl groups of glucose and the side chains of Thr194, Lys195, Asn229, Asp230, Asn256, Glu284 and Glu315, as well as an ordered water molecule within the cleft (Fig. AtHXK1 thus binds glucose in a mode quite similar to those of HXKs and GKs Aleshin, Zeng, Bourenkov et al, 1998;Kuettner et al, 2010;Lunin et al, 2004;Rosano et al, 1999). The residues of the glucose-binding site are conserved in all known HXKs and GKs except fructokinases (FRKs) ( Supplementary Fig.…”

“…EU247513), which was designated previously as HXK1 (Bluhm et al, 2008), was selected for characterization based on its expression profile during kernel colonization (Bluhm et al, 2008) and the similarity of the predicted Hxk1 protein to a functionally characterized hexokinase from Kluyveromyces lactis (KlHxk1, GenBank accession no. CAA08922.1; Kuettner et al, 2010). HXK1 has an ORF of 2329 bp with five introns, and is predicted to encode a protein of 493 aa containing two structurally similar domains.…”
