Crystallographic investigations of select cathinones: emerging illicit street drugs known as `bath salts'

Abstract: The name `bath salts', for an emerging class of synthetic cathinones, is derived from an attempt to evade prosecution and law enforcement. These are truly illicit drugs that have psychoactive CNS (central nervous system) stimulant effects and they have seen a rise in abuse as recreational drugs in the last few years since first having been seen in Japan in 2006. The ease of synthesis and modification of specific functional groups of the parent cathinone make these drugs particularly difficult to regulate. MDPV… Show more

“…This act spurned a need for clandestine labs to design new synthetic cathinones to be marketed and sold as “plant food” to continue legal sales into the United States. Unfortunately, these new synthetic cathinones are being developed and sold quicker than specialty laboratories can develop assays to detect them [6, 19, 22, 36-39]. It takes too long to develop legislation for outlawing new synthetic drugs.…”

“…Current research on the more prevalent bath salts including MDPV, Methylone, α-PVP, and Mephedrone have revealed increased abuse liability, pharmacokinetic metabolism profiles, pharmacodynamic effects, neurotoxicity, and urine laboratory assay development which will be discussed in detail [11-19]. …”

“…Single crystal X‐ray diffraction (SC‐XRD), a SWGDRUG category A technique, was chosen as the validation method as it has the power to detect molecules and their arrangement within a crystal lattice. Historically, microcrystalline tests have been developed without any conclusive evidence that the drug molecule was incorporated into the crystal lattice, however more recently it has been the customary way of establishing that microcrystals are forming out of a mix of drug and reagent solution, and indeed contain the drug molecule combined with the microcrystalline test reagent proving that the drug molecule as a whole is responsible for the interpretable result of the test, unlike solely targeted moieties as can be seen in colour tests . Indeed, the combination of several reagents in flow‐chart analysis style can lead to the identification of groups of compounds as the technique often only relates to a functional group rather than being specific for individual molecules …”
