Curcumin, Polydatin and Quercetin Synergistic Activity Protects from High-Glucose-Induced Inflammation and Oxidative Stress

Abstract: Chronic hyperglycemia, the diagnostic biomarker of Type 2 Diabetes Mellitus (T2DM), is a condition that fosters oxidative stress and proinflammatory signals, both involved in the promotion of cellular senescence. Senescent cells acquire a proinflammatory secretory phenotype, called SASP, exacerbating and perpetuating the detrimental effects of hyperglycemia. Bioactive compounds can exert antioxidant and anti-inflammatory properties. However, the synergistic anti-inflammatory and antioxidant effects of the most… Show more

“…76,77 Quercetin, one of the most representative polyphenols, has proven to have a role in age-related diseases, exhibits neuroprotective activity, prevents diabetes, protects cells from highglucose induced inflammation and increases C. elegans lifespan. 20,78,79 One of the mechanisms in which this fat reduction might be happening is through a diminution in the ingestion rate as we can observe in both the E. coli and pumping assays. The ingestion of fluorescent bacteria was increased under the obese conditions, except for the one supplemented with the extract.…”

“…The primary human endothelial cells obtained from the umbilical vein (HUVEC) were extensively investigated as a human model of endothelial cells to assess the effects of senolytic and senomorphic compounds [ 26 , 27 , 28 , 29 ]. We observed that 10 µg/mL of ZOE exerts anti-inflammatory activity on LPS-stimulated yHUVEC by reducing the expression levels of IL-1β, IL-8, MCP-1, TNF-α, and ICAM-1.…”

“…Poly was dissolved in 0.5% dimethysulfoxide (DMSO) and directly diluted in the medium to a concentration of 10 µM for further experiments. The concentration of Poly was determined based on the literatures 50 , 51 and our preliminary experiment (Supplementary Figs. 4 and 5 ).…”
