Background:Acute coronary syndrome (ACS) is closely related to unstable plaques and secondary thrombosis. The inflammatory cells in plaques and their inflammatory products may be the cause for plaque instability and ruptures. The study aimed to disclose the changes of inflammatory factors including serum intracellular adhesion molecule-1 (ICAM-1), chitinase-3-like protein 1 (YKL-40), and lipoprotein-associated phospholipase A2 (Lp-PLA2) in patients with ACS and its clinical significance.Methods:A total of 120 patients with coronary heart disease (CHD) were categorized into 2 groups: 69 with ACS and 51 with stable angina pectoris (SAP); 20 patients with chest pain and normal angiography served as a control group. The 120 patients with CHD were categorized into single-vessel disease group, double-vessel disease group, and three-vessel disease group based on the number of coronary artery stenosis. The severity of coronary artery stenosis was quantified based on coronary angiography using Gensini score. They were further divided into mild CHD group with its Gensini score <26 (n = 36), moderate CHD group with its Gensini score being 26–54 (n = 48) and severe CHD group with its Gensini score >54 (n = 36). Serum levels of ICAM-1, YKL-40, and Lp-PLA2 of different groups were determined by enzyme-linked immunosorbent assay. Correlation between ICAM-1, YKL-40, Lp-PLA2, and Gensini score was analyzed.Results:The levels of serum inflammatory factors ICAM-1, YKL-40, and Lp-PLA2 were significantly higher in the ACS group than those in control group and SAP group (all P < 0.05); and compared with control group, no significant difference was observed in terms of the serum ICAM-1, YKL-40, and Lp-PLA2 levels in the SAP group (P > 0.05).The levels of serum ICAM-1, YKL-40, and Lp-PLA2 were not significantly different among control group, single-vessel disease group, double-vessel disease group, and three-vessel disease group (all P > 0.05). The levels of serum ICAM-1, YKL-40, and Lp-PLA2 were not significantly different among control group, mild CHD group (Gensini score <26), moderate CHD group (Gensini score 26–54), and severe CHD group (Gensini score >54) (all P > 0.05).Conclusions:The serum levels of ICAM-1, YKL-40, and Lp-PLA2 were correlated with different clinical types of CHD, but not well correlated the severity and extent of artery stenosis, suggesting that ICAM-1, YKL-40, and Lp-PLA2 might be involved in occurrence of instability of atherosclerotic plaque, and might reflect the severity of CHD mostly through reflecting the plaque stability.

ÖZAmaç: Tavşan karotis arteri çevresine yaka yerleştirilmesi ateroskleroz gelişiminin en önemli basamaklarından biri olan intimal kalınlaşmaya neden olur. Bu çalışmada, yaka modelinde vWF ekspresyon düzeylerinin yedinci ve on dördüncü günde incelenmesi amaçlanmıştır. Yöntem: İnert, yumuşak silikon yaka sol karotis arter çevresine yedi veya on dört gün süre ile yerleştirilmiştir. vWF ekspresyonu RT-PCR yöntemi ile incelenmiştir. Her bir arterin intima/medya (indeks) oranları ölçülmüştür. Bulgular: Yedinci gün ile on dördüncü gün kıyaslandığında indeks değerinin on dör-düncü günde daha da arttığı görülmüştür. Yaka vWF ekspresyon düzeyinde artışa neden olmuştur. Yakanın neden olduğu vWF düzeylerindeki artış incelenen her iki günde de aynı düzeyde bulunmuştur. Sonuç: Elde ettiğimiz bulgular yaka modeli ile oluşan intimal kalınlaşmada vWF'ün önemini vurgulamaktadır. Anahtar kelimeler: vWF, yaka, intimal kalınlaşma, ateroskleroz ABSTRACTObjective: Placement of a soft silicone collar around the carotid arteries of rabbits induces intimal thickening which is considered as one of the crucial steps in the development of atherosclerosis. In this study we aimed to investigate vWF expression levels on days 7 and 14 after collar placement. Methods: A soft inert silicon collar was placed around the left carotid artery for 7 and 14 days. vWF expression was investigated by quantitave RT-PCR. In each artery (collar or sham) cross-sectional areas of intima, media were measured and intima/ media ratio was calculated. Results: Intimal thickening was more pronounced on day 14 compared to day 7. However the collar caused increase in vWF expression which was found to be at the same level on days 7 and 14. Conclusion: Our findings emphasize the importance of vWF in collar-induced intimal thickening.

Cardiovascular diseases (CVDs), especially the coronary arterial diseases (CADs), have become the main cause of death all around the world, attracting attentions from the whole society. Even though considerable progresses have been made for the treatment of CADs, many clinical challenges remain to be overcome. In particular, effective biomarkers for CADs need to be developed to facilitate the early diagnosis and thus early treatment of the disease. Recently, the dysregulation of microRNAs (miRNAs) has been found to be involved in the progression of multiple CADs, manifested as altered levels of miRNAs at different disease phases, suggesting that miRNAs may be capable of serving as promising biomarkers for CADs. Here, we attempt to evaluate the possibility of miRNAs as biomarkers for CADs and compare these markers with previously reported ones. In this review, we will summarize the basic concepts and advances for CADs and miRNAs, with a special emphasis on miRNAs in the progression of CADs.
