CXCL8 in Tumor Biology and Its Implications for Clinical Translation

Abstract: The chemokine CXCL8 has been found to play an important role in tumor progression in recent years. CXCL8 activates multiple intracellular signaling pathways by binding to its receptors (CXCR1/2), and plays dual pro-tumorigenic roles in the tumor microenvironment (TME) including directly promoting tumor survival and affecting components of TME to indirectly facilitate tumor progression, which include facilitating tumor cell proliferation and epithelial-to-mesenchymal transition (EMT), pro-angiogenesis, and inhi… Show more

“…CXCL8 also named as interleukin-8 (IL8), is a proinflammatory chemokine and mainly expressed by the epithelial cells and macrophages. Released CXCL8 could recruit the neutrophil aggregation in tissues/organs which suffered by injury, infection, or inflammation ( 18 , 19 ). Recently, the transcriptomic analysis of skin in patients with DFU revealed that the expression of CXCL8 was significant increased compared to the control ( 20 ).…”

“…LAMC2 promotes migration and invasion via EMT, which is dependent on TGF-β1 and ZEB1 (Zinc Finger E-Box Binding Homeobox 1) integrin [ 31 ]. CXCL8, also known as interleukin-8 (IL-8), is a prototypic chemokine belonging to the CXC family and is responsible for the recruitment and activation of neutrophils and granulocytes to the site of inflammation [ 32 ]. Recent studies have shown that CXCL8 is essential for tumor cells to acquire and maintain this aggressive phenotype.…”

“…These all interact with one another to play their role in tumor pathogenesis [6,7]. Interleukin-8 (IL8) is a proinflammatory cytokine produced by different cell types in response to tissue infection, inflammation or injury [8][9][10]. It is also associated with the development of different types of cancer like lung cancer, prostate cancer, breast cancer, colorectal cancer, etc.…”
