Cycle- and function-related changes in lectin binding to human endometrium: a histochemical study with pronase treatment

Abstract: Forty-eight endometrial biopsy specimens were obtained during a normal menstrual cycle, during pregnancy and from patients with dysfunctional bleeding. The specimens were examined for Peanut (PNA), Soybean (SBA), Vicia villosa (VVA), Phytohem- (PHA), Lens culinaris (LCA) and Concanavalin (succ. Con A) agglutinin binding. The study was performed on paraffin sections using the pronase digestion and either the peroxidase-antiperoxidase or the avidin-biotin-peroxidase method. Cycle-related changes of the PNA, SBA,… Show more

“…Finally, it should be noted that lectin binding to normal endometrium remained largely unaffected by the cycle-related changes reported earlier [6,16], although ConA did express intraluminal reactivity more frequently during the secretory, rather than during the proliferative, phase of the menstrual cycle.…”

“…In general, the literature on Con A suggests that a weak homogeneous staining occurs across the uterine surface, irrespective of the physiological state of the epithelium or type of probes applied (Aoki et al, 1989;Bychkov and Toto, 1987;Duncan et al, 1988;Klentzeris et al, 1991;Kupryjanczyk, 1989). Conversely, the reported decrease in the density of Con A staining across the surface of ''infertile'' versus ''normal fertile'' women (Klentzeris et al, 1991) supports the present findings that indicate a likely role for ␣-mannosyl and ␣-glucosyl residues in preparing a receptive epithelium for blastocyst attachment.…”

“…In women, SBA binding to the uterine surface coat across the menstrual cycle was highly variable (Aoki et al, 1989;Kupryjanczyk, 1989;Soderstrom, 1987). However, in one example SBA binding in women was reported to be absent around the early-luteal phase and strongly expressed at the late secretory stage (Jones et al, 1998) suggesting that ␣-D-Gal and ␣-D-GalNAc may play a role in preparing the endometrium for pregnancy as in the macaque (Anderson et al, 1990) rabbit (Bukers et al, 1990) and marmoset.…”
