Cyclic nucleotide phosphodiesterase of rat pancreatic islets. Effects of Ca2+, calmodulin and trifluoperazine

Abstract: Cyclic nucleotide phosphodiesterase activity towards cyclic AMP and cyclic GMP was studied in extracts of rat islets of Langerhans. Biphasic Eadie plots [Eadie (1942) J. Biol. Chem. 146, 85-93] were obtained with either substrate suggesting the presence of both 'high'- and 'low'-Km components. The apparent Km values were 6.2 +/- 0.5 (n = 8) microM and 103.4 +/- 13.5 (6) microM for cyclic AMP and 3.6 +/- 0.3 (12) microM and 61.4 +/- 7.5 (13) microM for cyclic GMP. With cyclic AMP as substrate, phosphodeisterase… Show more

“…There are 11 different families of PDEs representing 21 different gene products (Bender and Beavo, 2006;Conti, 2000). Use of specific pharmacological inhibitors of certain isoforms of PDE has inferred the presence of PDEs 3 and 4 and calcium sensitive PDEs in β cells (Parker et al, 1995;Shafiee-Nick et al, 1995;Sugden and Ashcroft, 1981). Inhibition of PDE3B in particular has demonstrated the firmest evidence for a PDE being implicated in inhibiting insulin secretion.…”

“…Earlier work showed islets to contain a cytosolic Ca-calmodulin activated PDE, with low and high K m components for both cAMP and cGMP and a particulate form, which appeared insensitive to Ca-calmodulin [53,54,55].…”

“…PDEs present in ␤-cells have been determined as total PDE activities from crude islet preparations and the presence of PDEs 3 and 4, and calcium-sensitive PDEs, in ␤-cells have (Sugden and Ashcroft 1981;Parker et al, 1995;Shafiee-Nick et al, 1995). One, in particular, PDE 3B, has been shown to be activated by IGF-1 (Zhao et al, 1997) and leptin (Zhao et al, 1998).…”
