Cysteine Prevents the Reduction in Keratin Synthesis Induced by Iron Deficiency in Human Keratinocytes

Abstract: L-cysteine is currently recognized as a conditionally essential sulphur amino acid. Besides contributing to many biological pathways, cysteine is a key component of the keratin protein by its ability to form disulfide bridges that confer strength and rigidity to the protein. In addition to cysteine, iron represents another critical factor in regulating keratins expression in epidermal tissues, as well as in hair follicle growth and maturation. By focusing on human keratinocytes, the aim of this study was to ev… Show more

“…After treatments, cells were washed and collected by scraping with PBS containing 1 mM EDTA and low-speed centrifugation. Cell pellets were then lysed at 4 °C for 30 min in a buffer containing 20 mM Tris–HCl, pH 7.4, 150 mM NaCl, 5 mM EDTA, 5% (v/v) glycerol, 10 mM NP-40 and protease inhibitor tablets (Roche)61. The supernatant fraction was obtained by centrifugation at 15,000 × g for 10 min at 4 °C and then stored at −80 °C.…”

“…Briefly, 1 mg of HCT 116 p53−/− whole cell lysate was incubated with 30 μl of protein A/G agarose beads coated with 5 μg of anti-rpL3 (Primm, Milan, Italy) or anti-CBS (Santa Cruz Biotechnology) at 4 °C for 12 h. The beads were washed and boiled in the SDS sample buffer. The eluted proteins were loaded on 12% SDS-PAGE and detected by western blotting as previously reported [37, 38]. Aliquots of protein samples (30 μg) were resolved by 12% SDS-gel electrophoresis and transferred into nitrocellulose filters.…”

“…Western blotting analysis was performed as previously reported48. The membranes were challenged with anti-rpL3 (Primm, Milan, Italy), anti-CBS, anti-NFκB, anti-IκB-α, anti-pIκB-α and anti-β-actin (Santa Cruz Biotechnology).…”
