Cytogenetic Damage of Human Lymphocytes in Humanized Mice Exposed to Neutrons and X Rays 24 h After Exposure

Abstract: Detonation of an improvised nuclear device highlights the need to understand the risk of mixed radiation exposure as prompt radiation exposure could produce significant neutron and gamma exposures. Although the neutron component may be a relatively small percentage of the total absorbed dose, the large relative biological effectiveness (RBE) can induce larger biological DNA damage and cell killing. The objective of this study was to use a hematopoietically humanized mouse model to measure chromosomal DNA damag… Show more

“…The nuclei were counter-stained with DAPI Vectashield® mounting medium (Vector Laboratories, Inc., Burlingame, CA). [ 74 , 78 ]. Scoring and analysis of micronuclei per binucleated cells were quantified using Zeiss fluorescence microscope (Axioplan 2; Carl Zeiss MicroImaging Inc., Thornwood, NY) × 10 air objective lens and Metafer MSearch platform.…”

“…Recent advances in animal modelling are beginning to deliver more sophisticated systems that recapitulate clinically relevant environments. The best examples of these are mice with humanised immune systems (Wang et al 2021), lung disease or cardiovascular disease. One possible approach to the important question of how can we either validate our models or show that they are not helpful is to investigate differences in gene expression profiles between rodent and human disease and only use models with demonstrable similarities (Seok et al 2013).…”

“…Unique radiation quality exposures under development, beyond X-ray/gamma high-dose rate models, include ultra-high-dose rate [Pannkuk et al, 2022], neutron [Xu et al, 2015;Laiakis et al, 2021;Wang et al, 2021], and long-term, low, and variable-dose-rate exposures [Garty et al, 2020;Pannkuk et al, 2021]. Because an investigation new drug may produce a significant neutron component during prompt radiation exposure, a broad energy spectrum neutron irradiator was created to simulate these exposure conditions.…”
