The 2017 American College of Neuropychopharmacology (ACNP) conference hosted a Study Group on 4 December 2017, Establishing best practice guidelines to improve the rigor, reproducibility, and transparency of the maternal immune activation (MIA) animal model of neurodevelopmental abnormalities. The goals of this session were to (a) evaluate the current literature and establish a consensus on best practices to be implemented in MIA studies, (b) identify remaining research gaps warranting additional data collection and lend to the development of evidence-based best practice design, and (c) inform the MIA research community of these findings. During this session, there was a detailed discussion on the importance of validating immunogen doses and standardizing the general design (e.g., species, immunogenic compound used, housing) of our MIA models both within and across laboratories. The consensus of the study group was that data does not currently exist to support specific evidence-based model selection or methodological recommendations due to lack of consistency in reporting, and that this issue extends to other inflammatory models of neurodevelopmental abnormalities. This launched a call to establish a reporting checklist focusing on validation, implementation, and transparency modeled on the ARRIVE Guidelines and CONSORT (scientific reporting guidelines for animal and clinical research, respectively). Here we provide a summary of the discussions in addition to a suggested checklist of reporting guidelines needed to improve the rigor and reproducibility of this valuable translational model, which can be adapted and applied to other animal models as well.

Depression is a major contributor to the global burden of disease and disability, yet it is poorly understood. Here we review data supporting a novel theoretical model for the biology of depression. In this model, a stressful life event leads to microdamage in the brain. This damage triggers an injury repair response consisting of a neuroinflammatory phase to clear cellular debris, and a spontaneous tissue regeneration phase involving neurotrophins and neurogenesis. During healing, released inflammatory mediators trigger sickness behavior and psychological pain via mechanisms similar to those that produce physical pain during wound healing. The depression remits if the neuronal injury repair process resolves successfully. Importantly, however, the acute psychological pain and neuroinflammation often transition to chronicity and develop into pathological depressive states. This hypothesis for depression explains substantially more data than alternative models, including why emerging data show that analgesic, anti-inflammatory, pro-neurogenic and pro-neurotrophic treatments have antidepressant effects. Thus, an acute depressive episode can be conceptualized as a normally self-limiting but highly error-prone process of recuperation from stress-triggered neuronal microdamage.

Immunomodulatory, anti-inflammatory and wound healing properties of Echinacea species

Herbal or botanical supplements have multiple health benefits. Public interest in these supplements has increased greatly in recent years aimed at modulation of the immune system to combat immune-related diseases. Historically, Echinacea has long been used as phytotherapy for wound healing, pain relief and treatment of the common cold. Although the non-specific immunostimulating properties of Echinacea species have been widely investigated, academic knowledge of the adaptive immune-modulating activity, the antiinflammatory activity and the mechanistic basis for these immunomodulatory properties remain elusive. In this dissertation, we focused our research interest in these less explored fields, with an emphasis on the herbal anti-inflammatory activity in both in vitro macrophage cell line and in vivo oral administration models. In vivo, alcohol extracts of Echinacea exhibited multiple immune-modulating effects. In addition to the non-specific, innate immunomodulation, they enhance B cell and T cell function (determined by increased production of antibodies and cytokines). These modulating effects of Echinacea were more robust when host immune functions were disturbed by mild stress. In vitro, alcohol extracts of Echinacea suppressed activated macrophages (RAW 264.7 cells) so they produced lower amounts of inflammatory mediators including nitric oxide (NO). The decreased NO production might be relative to decreased inducible nitric oxide synthase (iNOS) enzyme expression and increased arginase activity, suggesting that Echinacea could switch macrophage to alternate activation. In a cutaneous wound healing model, we demonstrated that alcohol extract of E. pallida helped improve restraint stress-delayed wound healing. These data provided novel evidence to support the multiple immunomodulatory properties of Echinacea and will help define the mechanisms behind the success of traditional use of Echinacea for pathogenic infections and inflammatory diseases.
