Cytokine MIF Enhances Blood-Brain Barrier Permeability: Impact for Therapy in Ischemic Stroke

Abstract: Ischemic stroke is a devastating disease with limited therapeutic options. It is very urgent to find a new target for drug development. Here we found that the blood level of MIF in ischemic stroke patients is upregulated. To figure out the pathological role of MIF in ischemic stroke, both in vitro and in vivo studies were conducted. For in vitro studies, primary cortical neuron cultures and adult rat brain endothelial cells (ARBECs) were subjected to oxygen-glucose deprivation (OGD)/reoxygenation. Middle cereb… Show more

“…It has been shown to promote leukocyte‐endothelial cell interactions through promoting endothelial adhesion molecule expression . It has recently been suggested to directly degrade the BBB after ischemic brain . Perivascular microglia/macrophages migrate toward the disrupted blood vessels and further damage them by phagocytizing ECs .…”

“…Besides, miR-186-5p is previously reported to serve as a circulating biomarker of cardiovascular risk and facilitate the diagnosis of acute coronary syndrome 6 , 15 . As reported, blood-brain barrier (BBB) was destroyed and its permeability was enhanced in ischemic stroke 16 , 17 . Thus, more small molecule materials can go through BBB and go into blood circulation.…”

“…The fact that ISO1 has been shown to influence astrocytes in animal models suggest that it can cross the blood brain barrier (BBB). Interestingly, MIF has been shown to increase BBB permeability and vascular leakage, whereas ISO-1 inhibits these effects [ 40 , 61 ]. Still, studies are needed to directly assess the ability of ISO1 to penetrate into the brain.…”

“…MIF inhibition has been shown to be neuroprotective in CNS disorders such as stroke [ 39 , 40 ], and MIF interaction with the CD74 receptor has been shown to activate astrocyte responses [ 41 ]. MIF-binding to CD74 can be inhibited by administration of the small molecule ISO1 [ 42 , 43 ].…”
