Cytokine secretion profile associated with periportal fibrosis in S. mansoni-infected Egyptian patients

Abstract: Periportal fibrosis (PPF) is a major pathological consequence of S. mansoni infection. Ultrasonography is a well-established tool for diagnosis and grading of schistosomiasis-related pathology. This work is performed to study the effect of schistosomiasis mansoni infection on the cytokine secretion profile in S. mansoni-infected patients at various grades of fibrosis, as determined by ultrasonography using Cairo Working Group classification. The levels of IL-2, IL-4, IL5, IL-10, IFN-gamma, and TNF-alpha were m… Show more

“…Human peripheral blood mononuclear cells (PBMCs) were obtained as described previously by Talaat et al . from heparinized blood by centrifugation through a Ficoll‐Hypaque separating media (Biowest SAS, Nuaillé, France) for 30 min in 24°C at 400 g (Sorvall centrifuge; Thermo Scientific, Pittsburgh, PA, USA). The PBMCs were collected from the interface and washed twice with RPMI‐1640 supplemented with L‐glutamine (200 mM), penicillin (100 U/ml), streptomycin (100 μg/ml) and 4‐(2‐hydroxyethyl)‐1‐piperazine ethanesulphonic acid (HEPES) buffer (1 M) (all from G ibco Life Technology Co., Carlsbad, CA, USA).…”

“…Anti-double stranded DNA (anti-dsDNA) antibody using a modified Farr assay. ml) in plasma of patients and controls were determined by ELISA as previously described [22,23] with slight modifications. A commercial DuoSet ELISA kits were used for detection of total concentrations of IL-6 (9.38-600 pg/ml), and TGF-b ( …”

“…The formation of granuloma in the proximity of schistosome eggs is mediated by T lymphocytes (47), and several studies have furnished insights into the cytokine cascade that controls the development of these lesions (7,10,45). We have recently evaluated the levels of production of the cytokines gamma interferon (IFN-␥), tumor necrosis factor alpha (TNF-␣) and TGF-␤, and interleukin-4 (IL-4), IL-10, and IL-13 following in vitro stimulation with soluble egg antigens (SEA) or soluble adult worm antigens of peripheral blood mononuclear cells (PBMC) from individuals of different ages and sexes and with various intensities of infection and degrees of fibrosis (2).…”
