Cytokines in Atherosclerosis: Pathogenic and Regulatory Pathways

Abstract: Atherosclerosis is a chronic disease of the arterial wall where both innate and adaptive immunoinflammatory mechanisms are involved. Inflammation is central at all stages of atherosclerosis. It is implicated in the formation of early fatty streaks, when the endothelium is activated and expresses chemokines and adhesion molecules leading to monocyte/lymphocyte recruitment and infiltration into the subendothelium. It also acts at the onset of adverse clinical vascular events, when activated cells within the plaq… Show more

“…20,21 IL-1b also influences the function of vessel wall elements, endothelial cells in particular and may contribute to the pathogenesis of arteriosclerosis in different ways, including by promoting coagulation and thrombosis. 22,23 Importantly, IL-1b was shown to control tumor angiogenesis and invasiveness of different tumor cells in mice. 24,25 Moreover, IL-1b plays a role in destructive joint and bone diseases.…”

“…In atherosclerosis, the activation of ECs in response to an inflammatory state leads to the expression and release of cytokines and chemokines, specifically interleukin (IL)-6 and monocyte chemoattractant protein (MCP)-1. These have been suggested to play the key role in the initial phase of atherosclerosis [20][21][22]. An increase in IL-6 levels has been associated with an increase in cardiovascular mortality in HIV-infected individuals [20], and MCP-1 has been shown to play a part in the attraction, migration, and activation of monocytes [23].…”

“…Les travaux chez l'animal et chez l'homme ont clairement montré que l'athérosclérose est une maladie inflammatoire chronique de la paroi des artères de gros et moyen calibre [3,4,37]. Les différentes étapes de l'athérogenèse comprennent la rétention et la modification oxydative des lipoprotéines dans l'espace sous-endothélial, l'activation et le dysfonctionnement des cellules endothéliales, puis l'adhésion, la migration et l'activation des leucocytes dans la paroi vasculaire.…”
