Cytolytic T-cell clones against an autologous human melanoma: specificity study and definition of three antigens by immunoselection.

Abstract: Cytolytic T-lymphocyte (CTL) clones against an autologous melanoma (SK-MEL-29) were generated by mixed lymphocyte tumor culture and subsequent cloning of responder lymphocytes at limiting dilutions. These CTL clones lysed autologous melanoma but not autologous Epstein-Barr virus-transformed B cells and none of the allogeneic tumor targets included in the specificity analysis. The lysis of autologous melanoma targets could be inhibited by monoclonal antibodies against monomorphic HLA class I determinants. For p… Show more

“…Previous studies have also depended on the establishment of long-term cultures of tumour cells which, in the context of developing a clinically useful cancer vaccine, is time-consuming and often inefficient, possibly resulting in the loss of critical tumour antigens. 28 This study describes the construction and application of a recombinant B7-1 adenovirus. This vector can rapidly and efficiently infect ovarian and cervical tumour cell lines with B7-1 expression detectable at the cell surface within 12 h following infection.…”

“…A single malignant cell may have multiple tumour-specific antigens (Wortzel et al, 1983;Knuth et al, 1989;Van den Eynde et al, 1989). Cytotoxic CD8 þ T cells recognise MHC class I molecules bearing antigenic peptides derived from endogenously synthesised proteins (Larsson et al, 2001).…”

“…The involvement of MHC class I antigens in the in vitro lysis of tumour cells by specific cytotoxic T-lymphocytes is well documented (Anichini et al, 1985;V'anky, 1986;Roberts et al, 1987;V'anky et al, 1987;Itoh et al, 1988;Darrow et al, 1989;Knuth et al, 1989).…”
