Cytoskeletal modulation of the response to mechanical stimulation in human vascular endothelial cells

Abstract: Possible interactions of cytoskeletal elements with mechanically induced membrane currents and Ca2+ signals were studied in human endothelial cells by using a combined patch-clamp and Fura II technique. For mechanical stimulation, cells were exposed to hypotonic solution (HTS). The concomitant cell swelling activates a Cl- current, releases Ca2+ from intracellular stores and activates Ca2+ influx. To interfere with the cytoskeleton, cells were loaded either with the F-actin-stabilizing agent phalloidin (10 mum… Show more

“…The similar results in other cells have been reported. In Blymphocyte, no I (Cl,swell) was recorded after F-actin integrity was disrupted by application of CB [9]. Sup-pression of RVD by CB was also observed in cultured PC12 cells [24], Necturus maculosus gallbladder epithelial cells [8] and isolated axons of the green crab Carcinus maenas [25].…”

“…Therefore, an intact F-actin network was considered to be critical for normal volume regulatory response in these cells. However, I (Cl,swell) in human endothelial cells was not affected by F-actin disruption [9], and F-actin disassembly in B-lymphocyte was not essential for activation of I (Cl,swell) [10]. So, the functional relationship between F-actin and I (Cl,swell) is still not clear.…”

“…Peptides-The peptide comprising the N-terminal 14 amino acids of bovine annexin II (STVHEILCKLSLEG) (25) and the L7E mutant version (STVHEIECKLSLEG) were synthesized on an automated sequencer (model 9050; Milligen). To verify the purity a small aliquot of the preparation was deprotected and subjected to automated sequence analysis.…”

“…2-4). Particularly, swelling-induced rearrangements of cytoskeletal elements regulate the opening of VRAC in some cell types (25)(26)(27), whereas a complete disruption of the cytoskeleton did not affect activation of I Cl,swell in endothelial cells (28).…”
