Cytosolic Phospholipase A2α and Eicosanoids Regulate Expression of Genes in Macrophages Involved in Host Defense and Inflammation

Abstract: The role of Group IVA cytosolic phospholipase A2 (cPLA2α) activation in regulating macrophage transcriptional responses to Candida albicans infection was investigated. cPLA2α releases arachidonic acid for the production of eicosanoids. In mouse resident peritoneal macrophages, prostacyclin, prostaglandin E2 and leukotriene C4 were produced within minutes of C. albicans addition before cyclooxygenase 2 expression. The production of TNFα was lower in C. albicans-stimulated cPLA2α+/+ than cPLA2α-/- macrophages du… Show more

“…infection has been reported recently by showing that PGE 2 promotes the formation of a complex between the actin depolymerization factor cofilin-1 and phosphatase and tensin homolog deleted on chromosome 10 that decreases polymerization of F-actin and increases monomeric G-actin (Serezani et al, 2012). A recent study disclosed how the timing of eicosanoid production may be coupled to either pro-and anti-inflammatory responses during Candida infection (Suram et al, 2013). Granulocyte-colony stimulating factor (csf3 gene), a cytokine that regulates the production and function of polymorphonuclear leukocytes, was the gene showing the highest degree of induction in response to Candida and its expression was reduced in cPLA 2 2/2 animals.…”

“…Expression-cPLA 2 ␣ initiates the release of arachidonic acid for production of prostaglandins and leukotrienes in RPM within minutes of infection with C. albicans (24). Prostaglandin I 2 (PGI 2 ) and prostaglandin E 2 (PGE 2 ) are produced before the expression of COX-2, suggesting that they are derived from the COX-1 pathway.…”

“…Prostaglandin I 2 (PGI 2 ) and prostaglandin E 2 (PGE 2 ) are produced before the expression of COX-2, suggesting that they are derived from the COX-1 pathway. By comparing C. albicans-infected cPLA 2 ␣ ϩ/ϩ and cPLA 2 ␣ Ϫ/Ϫ RPM, we previously found that cPLA 2 ␣-mediated production of prostaglandins regulates gene expression, including immediate early genes, such as Nr4a2, that are expressed within the first 30 -60 min of infection (24). To specifically investigate the role of COX-1-derived prostaglandins in regulating gene expression, RPM from wild type and COX-1 Ϫ/Ϫ mice were compared.…”
