Cytosolic Proteome Profiling of Aminoglycosides Resistant Mycobacterium tuberculosis Clinical Isolates Using MALDI-TOF/MS

Abstract: Emergence of extensively drug resistant tuberculosis (XDR-TB) is the consequence of the failure of second line TB treatment. Aminoglycosides are the important second line anti-TB drugs used to treat the multi drug resistant tuberculosis (MDR-TB). Main known mechanism of action of aminoglycosides is to inhibit the protein synthesis by inhibiting the normal functioning of ribosome. Primary target of aminoglycosides are the ribosomal RNA and its associated proteins. Various mechanisms have been proposed for amino… Show more

“…The CAO-specific integration site is located 338 bp upstream of Rv1360 and is in the same orientation. An overexpression of Rv1360 was determined in amikacin-and kanamycin-resistant M. tuberculosis isolates, speculatively suggesting its role in the compensation of drug resistance (18). Finally, it may be noted that a PPE family gene (PPE19, a core mycobacterial gene, conserved in mycobacterial strains) is located immediately downstream of Rv1360.…”

“…This may lead to the negligence of the strains identified at the "genus" level, thus underestimating the accuracy of MALDI-TOF MS in identifying anaerobes at the genus level. Despite these problems, MALDI-TOF MS is still widely used in bacterial identification and other fields such as strain typing [54], detection of virulence factors [55] and evaluation of drug resistance [56][57][58].…”

“…40 STRING analysis revealed that overexpressed proteins of intermediary metabolism and respiration, lipid metabolism, virulence, detoxification, adaptation, cell wall and cell processes, and conserved hypotheticals interacted with the other proteins, which were involved in various pathways of these categories. 20 We suggested that overexpressed proteins along with their interactome hold key pathways, which might cumulatively be involved in the pathogenesis of the prevalent strains of M. tuberculosis. In conclusion, comparative proteomic profiling of CF proteins of prevalent clinical M. tuberculosis isolates of north India by 2D-PAGE and MALDI-TOF/MS revealed consistently increased intensities of 12 proteins in all prevalent isolates, and of these, two are annotated with an unknown function.…”
