Ziziphus mucronata Willd. subsp. mucronata Willd. is a medicinal plant widely used in South Africa for the treatment of various diseases. Like many medicinal plants to which traditional claims including the treatment of ailments of infectious origin but without scientific reports have been associated, Z. mucronata is not an exemption. The ethanolic stems bark extract of Z. mucronata against medically important pathogens was investigated for the antimicrobial activity by the agar dilution method while the degree of its antimicrobial activity was determined by the macrobroth dilution method. The extract showed good antibacterial and antifungal activities. Both Gram-negative and Gram-positive bacteria were highly inhibited by the extract at very low concentrations. All the bacteria had inhibition zones greater than 20 mm at a concentration of 5 mg/ml with the exception of Pseudomonas aeruginosa (ATCC 19582) and Bacillus pumilus (ATCC 14884). For the bacteria, the minimum inhibitory concentration (MIC) values ranged between 0.078 and 0.625 mg/ml. For the fungi, the MIC values were between 2.5 to 10 mg/ml. The bacteria were more susceptible to the extract than the fungi. The antimicrobial activity of the extract indicated a broad spectrum, great therapeutic potential and justified the use of this plant by the rural communities in the treatment of microbial infections.

A sensitive, accurate, rapid and robust LC-MS-MS method for the quantification of aucubin, a major bioactive constituent of Aucuba japonica, Eucommia ulmoides and Plantago asiatica, was established and validated in rat plasma. Plasma samples were simply precipitated by adding methanol and the supernatant was chromatographed by a Diamonsil® C(18)(2) column with the mobile phase comprising a mixture of 10 mm ammonium acetate in methanol and that in water with the ratio of 50:50 (v/v). Quantification of aucubin was performed by mass spectrometry in the multiple-reaction monitoring mode with positive atmospheric ionization at m/z 364 → 149 for aucubin, and m/z 380 → 165 for catalpol (IS), respectively. The retention time was 2.47 and 2.44 min for aucubin and the IS, respectively. The calibration curve (10.0-30,000 ng/mL) was linear (r² > 0.99) and the lower limit of quantification was 10.0 ng/mL in the rat plasma sample. The method showed satisfactory results such as sensitivity, specificity, precision, accuracy, recovery, freeze-thaw and long-term stability. This simple LC-MS method was successfully applied in a pharmacokinetic study carried out in Sprague-Dawley rats after oral administration of aucubin at a single dose of 50 mg/kg. Herein the pharmacokinetic study of aucubin is reported for the first time.

Marsh hedge-nettle, Stachys palustris, is a perennial Eurasian member of the mint family, which has become naturalized and weedy in some parts of eastern North America. It is a hexaploid member of a holarctic species complex, which differs morphologically from the primarily tetraploid North American forms. The production of fleshy tuberous rhizomes is the most significant distinctive feature. In Canada, it has been a weed of potato and root crops in Nova Scotia, New Brunswick and Prince Edward Island, although other crops and areas, such as soybean in Ontario, are also affected. Usually considered a wetland plant, it readily spreads into drier arable fields from adjacent ditches and wetlands. Several herbicides available for use in Canada have been found to provide limited long-term suppression in agricultural systems, with the best results obtained using combined pre-emergent and post-emergent applications.
