Cytotoxic Activity of Aporphine Alkaloids from Alseodaphne Peduncularis (Wall. Ex. Ness) Meissn against Hela Cell Line

Abstract: Genus of Alseodaphne has been known as a source of apoprhine alkaloid that displayed potent cytotoxic activities against selected cell lines. Two aporphine alkaloids, Corydine 1 and norlirioferine 2 were isolated from leaves of Alseodaphne peduncularis (Wall. Ex. Ness) Meissn. (Lauraceae). Phytochemical study involved extraction, separation and purification by using various chromatography methods and structural elucidation by using spectoscopic techniques such as UV, IR LC-MS and 1D and 2D NMR. These compounds… Show more

“…Vincristine, a conventional drug of cancer, was used as a positive control against HeLa cell line and showed CD 50 value of 0.4 μg/ml. [24] The cytotoxicity of the extracts was assayed at various concentrations of 1000, 500, 100, 50, 20, 10, 5, and 1 μg/ml under continuous exposure for 72 h, which are expressed in CD 50 values and are summarized in Table 2.…”

“…Recently, Amna et al . () observed that laurotetanine showed very potent activity against heLa cells in vitro but none of the four aporphinoid alkaloids they tested had any effect against normal mouse fibroblast (NIH/3T3) cell lines according to the 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐ diphenyltetrazolium bromide (MTT) assay. Some aporphinoids also possess a vasodilator effect (by inhibition of extracellular calcium entry and sometimes tissue‐specific modifications of intracellular calcium movements) and anti‐platelet activity.…”

“…Anticancer activities in vivo have been reported in few cases and the dimeric molecule thaliblastine had been advanced to human clinical trials but these were halted due to no significant therapeutic activity being observed (St evigny et al 2005). Recently, Amna et al (2017) observed that laurotetanine showed very potent activity against heLa cells in vitro but none of the four aporphinoid alkaloids they tested had any effect against normal mouse fibroblast (NIH/3T3) cell lines according to the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Some aporphinoids also possess a vasodilator effect (by inhibition of extracellular calcium entry and sometimes tissue-specific modifications of intracellular calcium movements) and anti-platelet activity.…”
