Cytotoxic and Apoptotic Effects of Pinostilbene and Bortezomib Combination Treatment on Human Multiple Myeloma Cells

Anna Staskiewicz,
Erica Wong,
Michael Tucker
et al.

Abstract: Multiple myeloma (MM) is a cancer of plasma cells in the bone marrow characterized by bone lesions, hypercalcemia, anemia, and renal failure. Bortezomib (BTZ), a common treatment for MM, is a proteasome inhibitor that induces apoptosis in MM cells. However, high doses of BTZ can be very toxic, signifying a need for a synergistic drug combination to improve treatment efficacy. Resveratrol (RES), a phenolic compound found in grapes, has been shown to inhibit MM cell growth. We sought to identify a synergistic co… Show more

“…Apoptosis of RGD-IONP/CAPE treated RPMI8226 cells was examined using an Annexin V-FITC (AV)/7-Amino-Actinomycin (7-AAD) dual staining of cells followed by flow cytometry analysis as previously reported ( Staskiewicz et al, 2023 ). Cultured RPMI8226 cells (1.0 × 10 6 cells/well) were plated in 6-well plates and treated with RGD-IONP/CAPE at various concentrations (1, 5, 10, 20 and 30 µM), IONP/CAPE at 30 μM, CAPE only (30 µM), and RGD-IONP as control for 24 h. After treatment, cell staining was performed using a FITC Annexin V Apoptosis Detection Kit following the manufacturer’s protocol.…”
