Nowadays, cancer is one of the most common diseases in humans. Among all types, colorectal cancer (CRC) is one of the most serious types diagnosed in men after lung and prostate cancer while in women it occupies the second position after breast cancer worldwide. The risk factors such as obesity, sedentary lifestyle, bad nutritional habits (high in fats and proteins), smoking, and progressive aging are the cause of CRC. The acquisition of abnormal mutations leads to a consisting of many different arrangements of events during the tumor development process. Over the years, different approaches have been employed, in the treatment of cancer. These include chemotherapy, radiotherapy, surgery, and immunotherapy. Chemotherapy is routinely used for cancer treatment, but the toxicity of chemotherapeutics on healthy cells of the human body is obvious. This is the reason for discovering the new, natural origin, substances with potential cytostatic effects and less toxic side effects on the healthy cells. Medicinal plants have a special place in the management of cancer. Numerous cancer research studies have been conducted using traditional medicinal plants to discover new therapeutic agents with fewer side effects. In this review, we are describing two medicinal plants such as Actiniopteris radiata (Sw.) Link (Mayurashikha) and Terminalia pallida Brandis (Tella karaka) (endemic plant) which are available immensely in Chittoor District are used till today by the traditional herbal practitioners, tribal people is near to Talakona forest and Ayurvedic people for various diseases and also for CRC.

Objective: PITC-2 was isolated from the methanolic root extract of tissue cultured medicinal plant Pluchea indica (L.) Less. PITC-2 is a thiophene derivative which is 2-(Prop-1-ynyl)-5(5,6-dihydroxyhexa-1,3-diynyl)-thiophene. The main objective of the study is to evaluate the in vivo antitumor activity of PITC 2 against sarcoma-180 cancer cell in Swiss albino mice.Methods: The antitumor activity was evaluated by treatment with PITC-2 at a dose of 2.5 and 5 mg/kg b.w for 21 days on sarcoma-180 mice model. Cell viability was studied using 3-(4, 5- dimethylthiazol -2-yl)-2, 5-diphenyl tetrazolium bromide assay and cell apoptosis, G1 cell cycle arrest and reduction in tumor cell proliferation were evaluated by histopathological analysis and Bcl-2, cyclic-D1, and Ki-67 protein expression through immunohistochemistry study.Results: Precisely, PITC-2 had a cytotoxic effect on various in vitro cancer cells. Significant decreases in solid tumor volume and weight along with increase lifespan also observed. The histopathological and immunohistopathological examination indicates that PITC-2 induces apoptosis, typical morphological changes and suppresses tumor cell proliferation along with G1 cell cycle arrest through the downregulation of the intratumoral expression of Bcl-2, cyclic D1, and Ki-67 and thus highlighting antiproliferative and apoptotic properties against sarcoma-180 in vivo solid tumor model.Conclusion: The present results clearly demonstrate that PITC-2 significantly inhibits sarcoma-180 cell growth in a dose-dependent manner in in vivo mice model. Besides this, the study reveals a comprehensive perception of the possible mechanism behind the antitumor activity of PITC-2 by significant changes in the morphological, hematological, biochemical parameters in sarcoma-180 cells.

ABSTRAKPengembangan fitofarmakauntuk penyakit kanker sampai saat ini menjadi langkah utama dalam mengatasi kegagalan terapi kanker. Calotropis gigantea adalah salah satu tanaman yang telah terbukti secara ilmiah baik in vitro maupun in vivo sebagai agen kemopreventif. Penelitian ini bertujuan untuk menjelaskan efikasi dan keamanan ekstrak daun Calotropis gigantea (EDCG) dan ekstrak akar Calotropis gigantea (EACG) pada sel normal NIH3T3, sel kanker kolon WiDr dan sel kanker payudara T47D. Dari hasil analisis MTT dan analisis Selectivity Index (SI) didapatkan hasil bahwa Ekstrak etanol daun Calotropis gigantea (EDCG) dapat menghambat pertumbuhan sel kanker kolon WiDr secara selektif (SI>3) namun tidak selektif dalam menghambat pertumbuhan sel kanker payudara T47D (SI<3). Ekstrak etanol akar Calotropis gigantea (EACG) tidak selektif dalam menghambat pertumbuhan sel kanker kolon WiDr dan sel kanker payudara T47D (SI<3). Oleh karena itu EDCG dapat direkomendasikan sebagai kandidat Fitofarmaka dengan pembuktian lebih lajut melalui uji klinik.Katakunci : Calotropis gigantea, Sel NIH3T3, Sel WiDr, Sel T47D, efikasi, keamanan, Selectivity Index (SI) ABSTRACT Development of phytopharmaca for cancer until now has become a major step in overcoming the failure of cancer therapy. Calotrophis gigantea is one of the scientifically proven plants both in vitro and in vivo as a chemopreventive agent. This study aims to explain the efficacy and safety of Calotropis gigantea leaf extract (EDCG) and Calotropis gigantea root extract (EACG) on NIH3T3 cell, colon cancer cell WiDr and T47D breast cancer cells. From the analysis of MTT and Selectivity Index (SI) analysis showed that ethanol extract of leaf Calotropis gigantea (EDCG) can inhibit selective growth of selective colon cancer cells (SI> 3) but not selective in inhibiting the growth of breast cancer cells T47D (SI <3 ). Calotropic gigantea (EACG) root ethanol extract is not selective in inhibiting the growth of colon cancer cells WiDr and breast cancer cells T47D (SI <3). Therefore EDCG can be recommended as a Phytopharmaca candidate with further proof through clinical trials.
