Cytotoxic effect of diphtheria toxin used alone or in combination with other agents on human renal cell carcinoma cell lines

Abstract: Treatment of renal cell carcinoma (RCC) by conventional chemotherapy and immunotherapy has resulted in minimal remissions. Alternative forms of therapy are therefore being sought. The present study investigated the sensitivity of RCC cell lines to several toxins used alone and in combination with other agents. RCC lines were relatively sensitive to the direct cytotoxic effect of diphtheria toxin (DTX), Pseudomonas aeruginosa exotoxin A (PEA) and ricin. Furthermore, DTX in combination with tumor necrosis factor… Show more

“…This strengthens the argument that ceramide generation contributes to diphtheria toxin toxicity. Because diphtheria toxin also potentiates the cytotoxic effect of TNF-␣, which signals via ceramide, in renal cell carcinoma, 57 ovarian carcinoma, 58 and prostate cancer cells, 59 it has been suggested that the agents share the same pathway of cytotoxicity. 60 We propose that ceramide constitutes an element in the death-signaling cascade initiated by diphtheria toxin and DT 388 -GM-CSF.…”

“…Self-activating retroviral vectors may be useful for development of other suicide genes for cancer therapy. Many suicide genes, such as that for diphtheria toxin or ricin, do not require the presence of a prodrug and may be useful for the treatment of cancers or human immunodeficiency virus type 1 infection (4,13,19,32). However, the toxicity of these genes to the virus producer cells has prevented their development as therapeutic agents.…”

“…Adding TNF-to the existing system was in agreement with supporting evidence of some publications showing a synergistic effect in cell cytotoxicity mediated by TNF-and diphtheria toxin. This was shown on ovarian cancer cell lines-sensitive or resistant to both diphtheria toxin and TNF-and on renal cell carcinoma cell lines (Morimoto et al, 1991, Mizutani et al, 1994. Using a construct in which both TNF-and DTA expressions are driven by H19 tumor specific promoter, would overcome the dose limiting toxic effects of the systemic delivery of TNF-protein.…”
