Cytotoxic Effects of Activated Human Monocytes and Lymphocytes to Anti‐D‐treated Human Erythrocytes in Vitro

Abstract: Incubation of human monocytes, derived from peripheral blood, with cell-free supernatants from mixed lymphocyte cultures resulted in morphological and functional changes in the mature macrophages. Activation of monocyte-derived macrophages by these factors resulted in a significant increase in their capacity to lyse anti-D-treated human erythrocytes. The lytic activity of both normal and activated macrophages appeared to be independent of erythrophagocytosis. T lymphocytes activated by either allogeneic cells … Show more

“…The following variation in technique could be considered: pure monocytes as effector cells [6], enzyme treatment of target cells [24], addition of cytochalasin B to the effector cell-target cell mixture [8], determination of the osmotic fragility of the target cells in addition to lysis [7] and prior activation of effector cells by incubation with cell-free supernatants from mixed lymphocytic cultures [28], Spontaneous cell-mediated cytotoxicity as discussed by Leibold and Peter [21 ] should also be considered. Another possibility would be to care fully search for IgA antibodies.…”

“…Human monocytes and neutrophils are able to lyse a variety of antibody-sensitized target cells in vitro, including tumor cell lines and erythrocytes (RBC) (9,14,22,23,27,29,37). This has led to the suggestion that antibody-dependent cell-mediated cytotoxicity (ADCC) may be important in tumor surveillance (12, 15, 38) as well as in the pathogenesis of hemolytic anemias.…”
