The diabetic foot ulcer (DFU) are serious problems to diabetes and may be associated with late healing and septic manifestation, consequently result in amputation which is an extortionate incident. The innate repair receptor (IRR) is expressed by injured tissues and is activated by erythropoietin produced and released into damaged tissue. Activation of the IRR might provide benefit for diabetic wound healing. This study evaluated effect of a specific IRR agonist, ARA290, on skin wound repair. The aim of the present study was to evaluate the effect of topical application based on ARA290, in streptozotocin-induced diabetic incision wound models rats. The treatment was performed daily, until day 14 after wound induction. Wound closure was determined and the features of the repaired tissue were examined, including amount of collagen and protein content, biochemical parameters, antioxidant status and proinflammatory cytokines. The data confirmed wound healing activities via macroscopic, biochemical, immunofluorescent and molecular methods. There was meaningful acceleration in wound closure rate, decrease in the period of re-epitalization, greater amount of collagen and protein content in ARA290 treated group when compared with control group. The increase of serum insulin and HDL was divergent with blood glucose decrease and reduced lipid level. The healing effect was confirmed by reduced levels of inflammatory cytokines and lipid peroxidation and augmented antioxidants. The results propose that ARA290-arbitrated IRR activation may signify an appealing approach to treat diabetes-associated wound healing.

Background: Several studies have reported the incidence of gastric cancer (GC) in the patients with schizophrenia, but the incidence of GC in schizophrenia patients remains controversy. It is necessary to perform a meta-analysis to detect the incidence of GC in schizophrenia patients. Methods: We search the relevant articles from PubMed, EMBASE, Web of Science, the Cochrane Library and Chinese electronic databases. The incidence of GC in schizophrenia patients and heterogeneity were analyzed with STATA10.0 software. Results: Our meta-analysis included total 283,422 patients with schizophrenia and 1,024 patients with GC in nine articles. The incidence of GC in schizophrenia patients was 0.62% (95%CI: 0.005-0.008; P<0.001), but there is obvious heterogeneity (I 2 =98.7%). We next performed subgroup analysis to explore the incidence of GC in schizophrenia patients by gender and region. The GC incidence in male schizophrenia patients (0.28%, 95%CI: 0.001-0.003; P<0.001) was higher than that in female patients (0.23%, 95%CI: 0.002-0.004). In European countries, the GC incidence in schizophrenia patients (0.98%, 95%CI: 0.005-0.008) was higher than that in Asian countries (0.13%, 95%CI: 0.000-0.002; P<0.001). Conclusion: The incidence of GC in patients with schizophrenia was 0.62%. The GC incidence was higher in male and European countries schizophrenia patients, but large cohort studies are needed to confirm further.

Synthesis and In Vitro Evaluation of Novel Dopamine Receptor D2 3,4-dihydroquinolin-2(1H)-one Derivatives Related to Aripiprazole

In this pilot study, a series of new 3,4-dihydroquinolin-2(1H)-one derivatives as potential dopamine receptor D2 (D2R) modulators were synthesized and evaluated in vitro. The preliminary structure–activity relationship disclosed that compound 5e exhibited the highest D2R affinity among the newly synthesized compounds. In addition, 5e showed a very low cytotoxic profile and a high probability to cross the blood–brain barrier, which is important considering the observed affinity. However, molecular modelling simulation revealed completely different binding mode of 5e compared to USC-D301, which might be the culprit of the reduced affinity of 5e toward D2R in comparison with USC-D301.
